Ruim baan voor moderne veredelingsmethoden

Midden in de komkommertijd was het nog nooit zo spannend voor de biotech sector. Kunnen we straks gewassen ontwikkelen waar de samenleving om staat te springen? Gewassen die duurzamer geteeld kunnen worden, gezond en lekker zijn. Ligt moderne plantenveredeling nu echt binnen handbereik? Op 25 juli zal het Europese Hof van Justitie (EHJ) uitspraak doen of mutagenese en de moderne vormen daarvan uitgezonderd zijn van de wetgeving voor genetisch modificatie (GM-wetgeving). Advocaat Generaal Bobek heeft dit in zijn op 23 januari 2018 uitgebrachte advies al bevestigd. HollandBIO ziet dit als een grote kans en hoopt dat het Europese Hof het advies overneemt.

Ter voorbereiding van de EHJ uitspraak is er op 21 juni een projectbijeenkomst geweest met de betrokken leden. Daarin zijn de verschillende scenario’s van de uitspraak en de mogelijke gevolgen daarvan voor het bedrijfsleven besproken. We hopen natuurlijk dat ook het EHJ bevestigt dat de moderne veredelingsmethoden niet onder de GM-wetgeving vallen. Dat zou betekenen dat alle gewassen ontwikkeld met die methoden en waarin geen soort-vreemd DNA is geïntroduceerd (en die dus ook in de natuur kunnen voorkomen), niet onder de GM-wetgeving vallen. Aanvullende wetgeving is volgens ons ook niet nodig. De reguliere regelgeving voor milieu, zaden en planten en voedsel zijn voldoende om de veiligheid te waarborgen. Zie hiervoor ook het recent gepubliceerde artikel van een aantal juridische experts. Maar ook als het EJH anders besluit, zullen we de uitspraak aangrijpen om onze sector in een positief daglicht te zetten en momentum te creëren voor verandering.

HollandBIO is nu hard aan het werk om bovenstaande boodschap te verspreiden en uit te leggen. Daarvoor voert ze vele gesprekken met betrokken departementen, medestanders, de media en de politiek. Voor deze laatste groep is er in februari samen met Plantum een ‘De wereld draait door’ (DWDD) bijeenkomst georganiseerd. Tijdens die bijeenkomst konden de woordvoerders van de verschillende politieke partijen rechtstreeks in gesprek gaan met wetenschappers, bedrijven en milieuorganisaties. De bijeenkomst was zo succesvol dat er in het najaar een nieuwe DWDD-editie zal worden georganiseerd met de jongerenafdelingen van de politieke partijen.

Nieuw stimulerend biotech beleid

Een gezonde, duurzame en welvarende samenleving willen we allemaal. Maar tegelijkertijd staan we voor grote maatschappelijke uitdagingen, zoals een groeiende en vergrijzende wereldbevolking, een veranderend klimaat en een toenemende vraag naar duurzame productie van voedsel en energie. Bij het vinden van oplossingen is toepassing van (bio)technologie vaak doorslaggevend. Denk bijvoorbeeld aan katalysatoren in auto’s, zonnepanelen en windmolens, vaccins en geneesmiddelen.

HollandBIO maakt zich sterk voor een beleid dat het ontwikkelen en op de markt brengen van producten die een bijdrage leveren aan bovengenoemde maatschappelijke uitdagingen stimuleert. Daarin wordt biotechnologie gezien als een gangbaar bedrijfsproces waar de gangbare regelgeving van toepassing is en dat alleen waar nodig onder speciale regelgeving valt. We vinden dat het huidige, uitsluitend regulerende biotechnologiebeleid gebaseerd is op achterhaalde denkbeelden en pleiten daarom voor afschaffing. HollandBIO is daarom blij dat ook de Nederlandse overheid inziet dat het huidige biotechnologiebeleid aan modernisering toe is .

In juni 2017 is de modernisering van het biotech beleid op initiatief van het Ministerie van Infrastructuur en Watermanagement (IenW) van start gegaan met drie bijeenkomsten voor de medische, agri-food en industriële biotechnologie. Naast actieve deelname tijdens de bijeenkomsten, waar ook veel HollandBIO leden aanwezig waren, hebben we onze ideeën voor een stimulerend biotech beleid ook schriftelijk ingebracht.

Deze zomeractiviteiten werden in november 2017 afgesloten met een grote algemene bijeenkomst. Tijdens die bijeenkomst mocht Annemiek Verkamman een inleidende pitch geven om onze visie op een toekomstig biotech beleid te presenteren. Dit heeft ze vol verve gedaan. In de middagsessies werd er nader ingegaan op de vormgeving van een toekomstig beleid. Daar bleek dat de boodschap van Annemiek goed was overgekomen. Er was veel steun voor het, door HollandBIO aangeleverde, model. Binnen dat model wordt de ontwikkeling van producten die bijdragen aan oplossingen voor de maatschappelijke uitdagingen gestimuleerd en worden de kansen en mogelijke risico’s op een gebalanceerde manier tegen elkaar afgewogen (het innovatieprincipe). Begin 2018 heeft IenW de draad weer opgepakt met de stakeholders. Een brede Beraadsgroep en drie werkgroepen (medisch, agri-food en industrieel) zijn momenteel hard aan het werk om concrete bouwstenen aan te leveren voor een toekomstig biotech beleid. Ook binnen deze groepen worden onze ideeën voor een toekomstig biotech beleid goed ontvangen en gesteund. Doel voor de komende tijd is om onze wensen ook daadwerkelijk opgenomen te krijgen in het beleidsadvies van de werkgroepen aan het Ministerie van IenW. Vervolgens is het de bedoeling dat IenW het advies ook overneemt in haar beleidsvisie naar de Staatssecretaris en het Kabinet. En uiteindelijk: een biotech beleid waar onze leden wat aan hebben!

Strategic Collaboration Kite and Gadeta to Advance Gamma Delta T Cell Receptor Technology

Kite, a Gilead Company, and Gadeta, a privately-held company focused on the discovery and development of novel cancer immunotherapies based on gamma delta T cell receptors (TCRs), have entered into a strategic collaboration to develop novel gamma delta TCR therapies in various cancers. Under the financial terms, Kite will provide research and development (R&D) funding for the collaboration and Gadeta will be eligible to receive future payments upon achievement of certain regulatory milestones.  In addition, Kite will make an upfront purchase of equity in Gadeta from Gadeta’s shareholders and may acquire additional equity in Gadeta upon achievement of certain R&D milestones. Kite will have the exclusive option to acquire Gadeta.

Gadeta has developed a proprietary technology to engineer alpha beta T cells with gamma delta TCRs, called TEGs, for the potential treatment of various hematological cancers and solid tumors. This platform has the potential to combine the advantages of conventional T cells, which express alpha and beta TCR chains, with TCRs derived from gamma delta T cells that recognize novel targets in cancer cells, according to preclinical models evaluating the lead TEG candidates. Unlike alpha beta T cells, gamma delta TCRs do not require expression of cell surface proteins (major histocompatibility complex (MHC) molecules) for target recognition, and their ability to recognize novel targets under stress or metabolic conditions offer an attractive approach to develop potentially effective cell therapies in solid tumors.

Gadeta was founded in 2015 by Professor Jürgen Kuball, Mark de Boer, Utrecht Holdings and Medicxi, its founding investor.

Duidelijkheid over de bestrijding van kinkhoest, meningokokken en rotavirus

Met drie Kamerbrieven heeft staatssecretaris Blokhuis meer helderheid gegeven over nieuwe vaccinatiestrategieën in de bestrijding van kinkhoest, meningokokken en rotavirus. Een goede ontwikkeling, vindt HollandBIO. Zij roept op om de voorgestelde trajecten zo snel en breed mogelijk te implementeren. Vooral bij het rotavirusvaccin is daar wat HollandBIO betreft nog een punt van zorg.

Invoering maternale kinkhoestvaccinatie
Vanaf 2019 biedt de Jeugdgezondheidszorg (JGZ) (aanstaande) moeders de maternale kinkhoestvaccinatie aan. Om twee redenen vindt HollandBIO dit een uitstekende zet. In de eerste plaats omdat we hiermee jaarlijks ongeveer 100 ziekenhuisopnames en één sterfgeval ten gevolge van kinkhoest voorkomen.[1] Ten tweede omdat staatssecretaris Blokhuis hiermee slim gebruik maakt van de jarenlange ervaring van de JGZ met de voorlichting en uitvoering van het Rijksvaccinatieprogramma. Blokhuis kondigt in zijn brief ook gelijk een evaluatie van deze strategie aan na twee jaar.

Uitbreiding van de meningokokken ACWY vaccinatiestrategie
Staatssecretaris Blokhuis is -net als HollandBIO- bezorgd over het stijgend aantal meningokokken W infecties. Om deze stijging te beteugelen omarmt hij het recente advies van het “outbreak management team”, waarin zij voorstelt om naast baby’s van 14 maanden en kinderen van 14 jaar ook alle kinderen tot en met 18 jaar in 2019 een vaccinatie aan te bieden. Daarnaast stemt Blokhuis in met onderzoek naar dragerschap onder adolescenten en de werkzaamheid van deze vaccinatie bij 65-plussers. Het RIVM speelt bij beide maatregelen een sleutelrol, aangezien zij zowel de onderzoeksvoorstellen als de extra campagne uitwerkt.

Rotavirusvaccinatie enkel voor risicogroepen
De Staatssecretaris geeft vooralsnog voorkeur aan een risicogroepstrategie voor rotavirusvaccinatie. Hierdoor krijgen vanaf 2019 alleen vroeggeboren kinderen, kinderen met een te licht geboortegewicht en/of een aangeboren afwijking een vaccinatie aangeboden via het Rijksvaccinatieprogramma. De brief die de kers op de taart kon worden, is wat HollandBIO betreft de domper uit het rijtje: juist omdat de Gezondheidsraad zich positief uitsprak over algehele vaccinatie en de daaruit voortkomende groepsimmuniteit.

De staatssecretaris benoemt verschillende factoren die tot deze beslissing leiden. Hij haalt in de brief de conclusie van de Gezondheidsraad aan dat algehele vaccinatie niet kosteneffectief is, maar houdt daarbij geen rekening met het te verwachten inkoopvoordeel. De brief wijst echter ook op andere factoren die tot een conservatieve afweging van de staatssecretaris leiden. Die terughoudendheid komt vooral voort uit de angst dat uitbreiding van het Rijksvaccinatieprogramma in combinatie met een nieuwe toedieningsvorm en de toenemende twijfel over vaccinatie zouden leiden tot het niet halen van de grens voor groepsimmuniteit. HollandBIO is teleurgesteld dat argumenten op basis van angst en onbekendheid de doorslag geven in de afweging, terwijl deze te ondervangen zijn met goede voorlichting en uitvoering. Iets wat ook de  Gezondheidsraad stelt in haar advies.

HollandBIO vindt dat de aanzienlijke gezondheidswinst van algehele rotavirusvaccinatie doorslaggevend zou moeten zijn, juist omdat dit ook bij de risicogroepen tot meer gezondheidswinst leidt. Dit voorkomt naar schatting 1.930-3.389 ziekenhuisopnames en 5-6 sterfgevallen per jaar.[2] HollandBIO hoopt daarom dat de staatssecretaris deze beslissing wil heroverwegen, in plaats van een evaluatie twee jaar na gedeeltelijke invoering af te wachten. Alleen op die manier zetten we nu al in op optimale gezondheidswinst.

Dutch Heart Foundation, UMC Groningen and Pivot Park Screening Centre collaborate on heart failure

Scientists from UMC Groningen and Pivot Park Screening Centre have started a study with the objective to prevent one of the causes of heart failure. It is the first time that a protein related to protection of the heart muscle from stiffening, will be screened to discover a starting point for development of a new medicine. The study builds on the earlier work by cardiologist Van der Meer and his team. Pivot Park Screening Centre’s ultra High Throughput Screening facilities and expertise are used to discover matches between the protein and compounds. This collaboration is financed by Top Sector Life Sciences & Health (designated by the Dutch Ministry of Economic Affairs and Climate Policy in 2011) to stimulate Public-Private Partnerships and is supported by the Dutch Heart Foundation (Hartstichting).

“This study is of great importance, since heart failure caused by the stiffening of the cardiac muscle, is a severe disease, with no good options for treatment, ” tells Marina Senten, leader of Alliance, Science and Innovation at The Dutch Heart Foundation. “If we do nothing, more and more people will suffer from heart failure. Research on this topic is one of our top priorities. We therefore facilitated this Public-Private Partnership. This study reinforces our other studies on early detection and prevention of heart failure.”

During earlier studies van der Meer and his research group identified the protein that is capable to protect the cardiac muscle from stiffening. “Identifying this protein was the first step in this process. We are very pleased that we’re able to continue our study and start looking for compounds that have the ability to increase the protein’s activity”, tells van der Meer.

Pivot Park Screening Centre’s robotic facilities make it possible to discover matches between the protein and the 300.000 compounds that will be tested. “First we will develop an assay, or testing environment, making it possible to screen the protein in our robotic system. 300.000 compounds will be tested and re-tested to determine which compounds are the best activators for this protein,” tells Helma Rutjes, Chief Operations Officer at PPSC. When matching compounds are found, the process of optimization will start, resulting in tests on human cardiac cells.

Genmab Enters Strategic Partnership with Immatics to Discover and Develop Next Generation Bispecific Cancer Immunotherapies

Genmab has entered into a research collaboration and exclusive license agreement with privately owned Immatics Biotechnologies GmbH (Immatics), to discover and develop next-generation bispecific immunotherapies to target multiple cancer indications. The deal strengthens Genmab’s position in immuno-oncology by combining Genmab’s proprietary technologies and antibody know-how with Immatics’ XPRESIDENT® targets and T-cell receptor (TCR) capabilities.  Genmab will receive an exclusive license to three proprietary targets from Immatics, with an option to license up to two additional targets at predetermined economics. The companies will conduct joint research, funded by Genmab, on multiple antibody and/or TCR-based bispecific therapeutic product concepts.

Genmab may elect to progress any resulting product candidates, and will be responsible for development, manufacturing and worldwide commercialization. For any products that are commercialized by Genmab, Immatics will have an option to limited co-promotion efforts in selected countries in the EU.

“This collaboration with Immatics gives us the opportunity to combine our unique technologies and expertise to create differentiated novel next-generation therapies. Under the terms of the agreement, Genmab will pay Immatics an upfront fee of USD 54 million and Immatics is eligible to receive up to USD 550 million in development, regulatory and commercial milestone payments for each product, as well as tiered royalties on net sales.

Biogen en overheid zijn het eens: SMA patiënten krijgen Spinraza

Biogen en de Nederlandse overheid hebben een overeenkomst bereikt over de vergoeding van Spinraza. HollandBIO is daar heel blij mee. Spinraza is het eerste en enige goedgekeurde medicijn voor de behandeling van spinale musculaire atrofie (SMA), een zeer ernstige, progressieve spierziekte. HollandBIO is er trots op een sector te mogen vertegenwoordigen die deze innovaties voortbrengt.

Door de overeenkomst komt het middel Spinraza voor circa 80 jonge kinderen met SMA vanaf 1 augustus in het basispakket. Dit is geweldig nieuws voor patiënten en hun ouders. Dorota Mazurkiewicz, Managing Director van Biogen Nederland licht de overeenkomst toe:

“Allereerst is dit prachtig nieuws voor de betreffende SMA-patiënten en hun families. SMA is een verschrikkelijke ziekte waar geen medicijn voor bestond, maar nu kunnen we patiënten nieuwe hoop bieden. We gaan nu met betrokken partijen in gesprek om de mogelijkheden voor voorwaardelijke vergoeding te verkennen. We hopen daarmee brede toegang voor alle SMA-patiënten te kunnen realiseren, ongeacht leeftijd of ziektetype, zoals ook het geval is in veel andere landen.”

We zijn er dus nog niet. Het middel betekent een kentering in het leven van patiënten. HollandBIO hoopt daarom dat alle patiënten met SMA zo snel mogelijk toegang krijgen tot Spinraza.

Galapagos announces design for PINTA Phase 2 trial with GLPG1205 in IPF

Galapagos announces the PINTA Phase 2 trial design with its GPR84 inhibitor GLPG1205 in patients with idiopathic pulmonary fibrosis (IPF). PINTA is a randomized, double-blind, placebo-controlled trial investigating a 100 mg once-daily oral dose of GLPG1205. The drug candidate or placebo will be administered for 26 weeks in up to 60 IPF patients. Patients may remain on their local standard of care as background therapy. Primary objective of the trial is to assess the change from baseline in Forced Vital Capacity (FVC in mL) over 26 weeks compared to placebo. Secondary measures include safety, tolerability, pharmacokinetics and pharmacodynamics, time to major events, changes in functional exercise capacity, and quality of life. IPF diagnosis will be confirmed by central reading. Recruitment for PINTA is planned in 10 countries in Europe, North Africa, and the Middle East. First dosing of an IPF patient is expected in the second half of 2018.

GLPG1205 is a GPR84 inhibitor discovered by Galapagos and fully proprietary to Galapagos. GLPG1205 showed a reduction in signs and symptoms in IPF animal models and has shown favorable tolerability in healthy volunteers and ulcerative colitis patients in previous trials. Galapagos currently has three drug candidates with distinct mechanisms of action in its fully proprietary portfolio aimed at building an IPF franchise: GLPG1690 in the ISABELA Phase 3 program, GLPG1205 in PINTA Phase 2, and GLPG3499, currently in pre-clinical development.

“GLPG1205 has shown signs of good activity in relevant animal models, and GPR84 has already been validated as a mechanism in combination with nintedanib[1] in IPF,” added Dr. Piet Wigerinck, Chief Scientific Officer of Galapagos. “We have a well-designed trial with PINTA for ‘1205 that we anticipate will give us new insights into the potential value of GPR84 inhibition as a mechanism to treat this highly fatal disease.”

First cohort enrolled in trial with innovative gene therapy for arthritis

Arthrogen, a clinical stage gene therapy company together with the Centre for Human Drug Research (CHDR), announced that the first three arthritis patients have been enrolled in a phase Ib gene therapy trial for treatment of arthritis with ART-I02.

ART-I02 is an adeno-associated virus (AAV5) vector encoding the human IFN-β gene under control of an inflammation-responsive promoter. ART-I02 is designed to produce the anti-inflammatory protein IFNβ in the synovial cells in the joint. The aim is to achieve a sustained clinical remission with a single treatment. This current clinical phase Ib trial, evaluates the safety, tolerability, pharmacokinetics, immunogenicity and anti-inflammatory activity of ART-I02 treatment in patients with rheumatoid arthritis (RA) or osteoarthritis (OA). In total 12 patients with RA or OA in their hand joints will be included. Parallel to this study, a similar phase Ib trial has started in Canada treating 15 patients with RA in their wrist.

“Patients with rheumatoid arthritis and osteoarthritis face chronic pain and immobility, even despite the availability of new, mostly systemic treatments.”, explains principal investigator Prof. dr. Koos Burggraaf of CHDR and he continuous: “In this trial we evaluate the safety of ART-I02 and we assess effect measures, hoping that this first step may prove to be an important milestone marking a new era in the treatment of arthritis, a chronic debilitating disease.”.
