Vakjury over IV-Walk

Facio Therapies has announced that oral treatment with one of Facio’s lead candidates results in significant reduction of the human muscle-toxic DUX4 protein in mice engrafted with human FSHD-affected muscle cells. Facio is the first in the FSHD field to achieve proof of principle in an animal model (“in vivo”).
The key event in the progressive muscle wasting disease FSHD is the undue production of the DUX4 protein in skeletal muscle. DUX4 sets in motion a cascade of biochemical events that eventually result in the devastating effects of FSHD. In people without FSHD, the production of DUX4 in skeletal muscle is repressed by regulatory mechanisms. Facio’s single goal is to develop therapies that restore this repression as much as possible.
Together with its drug discovery partner, Evotec, Facio successfully developed a stable mouse model based on engrafting human FSHD-affected muscle precursor cells (called “myoblasts”) onto a mouse thigh muscle. These human FSHD myoblasts then fuse and develop into mature muscle cells (called “myotubes”), which produce DUX4. Facio’s mouse model represents an important improvement over similar models, which either rely on engrafting human myotubes with low throughput, or on engrafting so-called “immortalized” FSHD cells that deviate from natural FSHD biology because they have been altered to remain alive indefinitely. Facio, however, approximates natural FSHD biology as much as possible by using unadulterated (or “primary”) FSHD-affected muscle cells in its mouse model. Primary FSHD muscle cells are also the basis of Facio’s unique high-throughput DUX4 screening platform.
“Achieving stability was the main challenge”, said Facio’s Managing Director, David Dasberg, “but after about six months we were able to stably engraft human FSHD myoblasts in one mouse thigh and healthy human myoblasts in the other thigh, so that each mouse serves as its own control. DUX4 expression does occur in FSHD myotubes but not in healthy myotubes, which validates the model. Then, in a single-concentration experiment, we orally treated mice with one of our lead candidates or with a placebo. This resulted in a statistically highly significant reduction of DUX4 and of certain DUX4 target genes in human FSHD-affected myotubes. Evidencing that an orally administered compound is able to suppress human DUX4 in a representative animal model is a major leap forward in developing an FSHD therapy. We are extremely pleased with these results, all the more so because chemical optimization of our lead candidates for specific use in FSHD will further improve their efficiency at reducing DUX4.”
Facio has begun expanding its in vivo proof of principle by including other lead candidates, by performing dose-response experiments, and by studying the effect of lead candidates on human muscle cell formation. Together with lead optimization, this will enable selecting two lead compounds for government-mandated pre-clinical safety studies. The lead compound with the best overall performance in terms of efficacy and safety will be taken into a first-in-human clinical trial. As announced previously, Facio currently plans to start a first-in-human trial in the second half of 2021. In consultation with leading clinical experts, Facio is developing an integrated regulatory and clinical development strategy specific to FSHD. “That strategy is the last barrier on the way to clinical trials,” explained David. “We are confident we will cross that barrier, just as we successfully crossed the barrier of establishing a primary-cell-based DUX4 screening platform and then the barrier of demonstrating in vivo proof of principle.”
David stressed that in vivo data represent a key milestone along the way to clinical trials. “It has been said that an FSHD-relevant animal model is technically impossible. We now show that this assertion is incorrect. This has important consequences for the preparation of clinical studies. It is risky to jump from the relatively simple reality of a dish (“in vitro”) straight into the highly complex reality of people with FSHD. After all, results achieved in a dish say nothing a compound’s ability to enter FSHD-affected muscle, or about the dose required to reach the desired effect in such muscle. High doses increase the risk of side effects, especially because an FSHD therapy will be chronic. These risks are mitigated by in vivo proof-of-principle data. Such data should be seen as a crucial component of a regulatory submission for the initiation of a first-in-human study (known as an IND in the US, and as an IMPD in Europe).”
Kees van der Graaf, Facio’s Chairman, noted: “Only two years ago, we were the first to enable reliable quantification of the DUX4 protein in primary FSHD muscle cells. In fact, we still are the only one in the FSHD field with a fully automated, high-throughput DUX4 screening platform. Now, we are the first in the FSHD field to achieve in vivo proof of principle. This double first demonstrates the power of our single focus on FSHD and our commitment to rigorous R&D. People with FSHD need real progress, and real progress requires rigor. We fully intend to stay the course as we prepare for clinical studies.”

Two Netherlands-based start-ups take major technological step in precision plant breeding, by showing that the SuRE technology can be successfully applied to the tomato genome, delivering a new genetic target identification platform for use in agricultural crops.
SuRE, ‘Survey of Regulatory Elements’, is a highly novel platform technology that allows unbiased genome-wide identification of gene regulatory elements that can serve as targets for mutagenesis. Through SuRE, we can identify unique and proprietary (patentable) targets that we can modify to yield new traits. SuRE outperforms competing technologies by a wide margin and has valuable agricultural applications, such as the identification of novel, unique and sophisticated targets for molecular plant breeding through targeted mutagenesis approaches. The technology is patent pending and was invented by Gen-X’s founder and published in the journal Nature Biotechnology, in 2017. HRB has partnered with Gen-X to translate the SuRE platform technology from validated pharmaceutical applications to new agricultural applications. These include the identification of novel, unique and sophisticated targets for molecular plant breeding through targeted mutagenesis approaches (TILLING and other gene-editing methods like CRISPR), as well as of novel, strong endogenous promoters to drive cis- and transgene expression.
Proof of Concept for SuRE in plants With SuRE, a plasmid library is constructed, consisting of random genomic fragments inserted upstream of unique 20-bp barcodes. The library is transfected into protoplasts, and barcode expression is quantified by high-throughput sequencing. Over 50-fold genome coverage can be reached, allowing robust mapping of autonomous promoter and enhancer activity to a genome.
In a project supported by Syngenta, Vegetable Seeds from BASF (Nunhems Netherlands), HZPC Research BV and Genetwister, HRB and Gen-X were able to show that the SuRE platform technology can be successfully applied to the tomato genome. The figure above shows that the SuRE library vector can be used to assay transcriptional regulation in tomato, and that when tomato genomic DNA is analyzed, a clear peak pattern is observed, representing gene regulatory elements.
“We are excited about achieving this major technical milestone. The combination of the proprietary SuRE and CRISPR technology platforms offers a powerful and unique way for plant breeders to improve crop yields and enable more sustainable food production” says Rudi Ariaans, CEO and co-founder of HRB.

ProQR receives exclusive worldwide license for IONIS-RHO-2.5Rx, now QR-1123, for autosomal dominant retinitis pigmentosa (adRP), a rare inherited form of blindness with no approved therapy. A first in human Phase 1/2 clinical trial in adRP patients is expected to start in 2019.
ProQR Therapeutics a company dedicated to changing lives through the creation of transformative RNA medicines for the treatment of severe genetic rare diseases, today announced the signing of an agreement with Ionis Pharmaceuticals to license QR-1123 (formerly “IONIS-RHO-2.5Rx”), an RNA medicine for autosomal dominant retinitis pigmentosa (adRP) caused by the P23H mutation in the rhodopsin (RHO) gene.
“Building on the recent proof of concept with QR-110 in LCA10 patients, we have confidence in the potential of single stranded RNA-targeted therapies to treat patients with genetic eye diseases,” said Daniel A. de Boer, chief executive officer of ProQR. “By acquiring the rights to QR-1123, we have strategically expanded our pipeline in retinal diseases. Ionis has a wealth of experience in discovery and development of antisense drugs and we look forward to working with them on this important program. Unlike other molecules we are developing, QR-1123 is a gapmer with a mutant allele-specific knockdown mechanism of action. If validated, it would further broaden the potential of RNA-targeted therapies in retinal diseases. We are excited to start clinical development to explore QR-1123’s potential in helping patients with P23H adRP.”
Under the terms of the agreement, ProQR was granted an exclusive worldwide license to QR-1123 and relevant patents. ProQR made an upfront payment in ordinary shares in the aggregate amount of $2.5 million, at $22.23 per share, which represents a 20% premium (based on the volume weighted average price of the previous 20 trading days) to its common stock, to Ionis upon signing the agreement. ProQR will also make future milestone payments, certain of which will be made in equity and others in cash or equity at ProQR’s discretion, and royalties on net sales of 20% through the royalty term.
Preclinical activities have been completed by Ionis and were presented at the 2015 annual meeting of the Association for Research in Vision and Ophthalmology (ARVO) and published in a peer-reviewed article, Murray et al 2015, IOVS. A natural history study in patients with P23H adRP has been conducted. The results of this study provide valuable information on disease progression in these patients, which is important data for designing the clinical development plan. ProQR expects to start a Phase 1/2 clinical trial in patients with adRP in 2019, pending submission and clearance of the IND application by the U.S. Food and Drug Administration (FDA).
Brett P. Monia, Ph.D., chief operating officer and senior vice president of translational medicine at Ionis, added, “IONIS-RHO-2.5Rx, now QR-1123, is an antisense oligonucleotide we designed to specifically target only the mRNA from the disease-causing rhodopsin gene, which has a single nucleotide P23H mutation, while preserving expression of the mRNA from the normal rhodopsin gene, which is important for the eye to function properly. IONIS-RHO-2.5Rx was developed by Ionis scientists dedicated to discovering innovative therapeutics to fight diseases where no other treatments have proven effective or even existed. We believe that ProQR has the ophthalmology expertise and experience in developing oligonucleotide drugs for the eye to rapidly bring this program through clinical development and ultimately to patients.”

Vakjury over IV-Walk

Facio achieves first-ever in vivo proof of principle in FSHD
GezondheidFacio Therapies has announced that oral treatment with one of Facio’s lead candidates results in significant reduction of the human muscle-toxic DUX4 protein in mice engrafted with human FSHD-affected muscle cells.

Hudson River Biotechnology and Gen-X deliver Proof of Concept for SuRE technology in plants
Voedsel en materialenTwo Netherlands-based start-ups take major technological step in precision plant breeding, by showing that the SuRE technology can be successfully applied to the tomato genome, delivering a new genetic target identification platform for use in agricultural crops.

ProQR In-licenses Worldwide Rights to Ophthalmology Drug Candidate from Ionis Pharmaceuticals
InnovatieklimaatProQR receives exclusive worldwide license for IONIS-RHO-2.5Rx, now QR-1123, for autosomal dominant retinitis pigmentosa (adRP), a rare inherited form of blindness with no approved therapy.
