ProQR raises $57.5 million

ProQR Therapeutics, a company dedicated to changing lives through the creation of transformative RNA medicines for severe genetic rare diseases, today announced the closing of its previously announced underwritten public offering of 9,090,909 ordinary shares at a price to the public of $5.50 per share.  All of the shares are being offered by ProQR.  The closing included the full exercise of the underwriters’ option to purchase up to 1,363,636 additional ordinary shares at the public offering price, less underwriting discounts and commissions.  Gross proceeds from the offering totaled approximately $57.5 million, before deducting underwriting discounts and commissions and offering expenses. 

Citigroup Global Markets and Evercore ISI acted as joint bookrunners for the offering.  Cantor Fitzgerald & Co. and JMP Securities acted as lead managers, with Chardan and Kempen acting as co-managers.

Biogen plans regulatory filing for aducanumab in Alzheimer’s disease

Biogen plans to pursue regulatory approval for aducanumab, an investigational treatment for early Alzheimer’s disease (AD), after consulting with the U.S. Food and Drug Administration (FDA). The Phase 3 EMERGE Study met its primary endpoint showing a significant reduction in clinical decline, and Biogen believes that results from a subset of patients in the Phase 3 ENGAGE Study who received sufficient exposure to high dose aducanumab support the findings from EMERGE. Patients who received aducanumab experienced significant benefits on measures of cognition and function such as memory, orientation, and language. Patients also experienced benefits on activities of daily living including conducting personal finances, performing household chores such as cleaning, shopping, and doing laundry, and independently traveling out of the home. If approved, aducanumab would become the first therapy to reduce the clinical decline of Alzheimer’s disease and would also be the first therapy to demonstrate that removing amyloid beta resulted in better clinical outcomes.

The decision to file is based on a new analysis, conducted by Biogen in consultation with the FDA, of a larger dataset from the Phase 3 clinical studies that were discontinued in March 2019 following a futility analysis. This new analysis of a larger dataset that includes additional data that became available after the pre-specified futility analysis shows that aducanumab is pharmacologically and clinically active as determined by dose-dependent effects in reducing brain amyloid and in reducing clinical decline as assessed by the pre-specified primary endpoint Clinical Dementia Rating-Sum of Boxes (CDR-SB). In both studies, the safety and tolerability profile of aducanumab was consistent with prior studies of aducanumab.

“With such a devastating disease that affects tens of millions worldwide, today’s announcement is truly heartening in the fight against Alzheimer’s. This is the result of groundbreaking research and is a testament to Biogen’s steadfast determination to follow the science and do the right thing for patients,” said Michel Vounatsos, Chief Executive Officer at Biogen. “We are hopeful about the prospect of offering patients the first therapy to reduce the clinical decline of Alzheimer’s disease and the potential implication of these results for similar approaches targeting amyloid beta.”

Based on discussions with the FDA, the Company plans to file a Biologics License Application (BLA) in early 2020 and will continue dialogue with regulatory authorities in international markets including Europe and Japan. The BLA submission will include data from the Phase 1/1b studies as well as the complete set of data from the Phase 3 studies.

The Company aims to offer access to aducanumab to eligible patients previously enrolled in the Phase 3 studies, the long-term extension study for the Phase 1b PRIME study, and the EVOLVE safety study. Biogen will work towards this goal with regulatory authorities and principal investigators with a sense of urgency.

Study Results
EMERGE (1,638 patients) and ENGAGE (1,647 patients) were Phase 3 multicenter, randomized, double-blind, placebo-controlled, parallel-group studies designed to evaluate the efficacy and safety of two dosing regimens of aducanumab. These studies were discontinued on March 21, 2019, following the results of a pre-specified futility analysis which relied on an earlier and smaller dataset. The futility analysis was based on data available as of December 26, 2018, from 1,748 patients who had the opportunity to complete the 18-month study period and predicted that both studies were unlikely to meet their primary endpoint upon completion. Futility analyses are common in large clinical studies and use statistical modeling to attempt to predict the outcome of the studies based on a number of pre-specified assumptions and criteria.

Following the discontinuation of EMERGE and ENGAGE, additional data from these studies became available resulting in a larger dataset, which included a total of 3,285 patients, 2,066 of whom had the opportunity to complete the full 18 months of treatment. A new extensive analysis of this larger dataset showed a different outcome than the outcome predicted by the futility analysis. Specifically, the new analysis of this larger dataset showed EMERGE to be statistically significant on the pre-specified primary endpoint (P=0.01). Biogen believes that data from a subset of ENGAGE support the findings from EMERGE, though ENGAGE did not meet its primary endpoint. Biogen consulted with external advisors and the FDA on these different results and their implications.

“This large dataset represents the first time a Phase 3 study has demonstrated that clearance of aggregated amyloid beta can reduce the clinical decline of Alzheimer’s disease, providing new hope for the medical community, the patients, and their families,” said Dr. Anton Porsteinsson, William B. and Sheila Konar Professor of Psychiatry, Neurology and Neuroscience, director of the University of Rochester Alzheimer’s Disease Care, Research and Education Program (AD-CARE), and principal investigator. “There is tremendous unmet medical need, and the Alzheimer’s disease community has been waiting for this moment. I commend Biogen, the FDA, the medical community, and the patients and their study partners for their persistence in working to make today’s announcement a reality.”

In EMERGE, which met its pre-specified primary endpoint in the new analysis, patients treated with high dose aducanumab showed a significant reduction of clinical decline from baseline in CDR-SB scores at 78 weeks (23% versus placebo, P=0.01). In EMERGE, patients treated with high dose aducanumab also showed a consistent reduction of clinical decline as measured by the pre-specified secondary endpoints: the Mini-Mental State Examination (MMSE; 15% versus placebo, P=0.06), the AD Assessment Scale-Cognitive Subscale 13 Items (ADAS-Cog 13; 27% versus placebo, P=0.01), and the AD Cooperative Study-Activities of Daily Living Inventory Mild Cognitive Impairment Version (ADCS-ADL-MCI; 40% versus placebo, P=0.001). Imaging of amyloid plaque deposition in EMERGE demonstrated that amyloid plaque burden was reduced with low and high dose aducanumab compared to placebo at 26 and 78 weeks (P<0.001). Additional biomarker data of tau levels in the cerebrospinal fluid supported these clinical findings. Biogen believes that data from patients in ENGAGE who achieved sufficient exposure to high dose aducanumab supported the findings of EMERGE.

In both studies, the most commonly reported adverse events were amyloid-related imaging abnormalities-edema (ARIA-E) and headache. The majority of patients with ARIA-E did not experience symptoms during the ARIA-E episode, and ARIA-E episodes generally resolved within 4 to 16 weeks, typically without long-term clinical sequelae. Biogen plans to present further detail on the new analysis of the larger dataset from EMERGE and ENGAGE at the Clinical Trials on Alzheimer’s Disease (CTAD) meeting in December 2019.

After reviewing the data in consultation with the FDA, Biogen believes that the difference between the results of the new analysis of the larger dataset and the outcome predicted by the futility analysis was largely due to patients’ greater exposure to high dose aducanumab. Multiple factors contributed to the greater exposure to aducanumab in the new analysis of the larger dataset, including data on a greater number of patients, a longer average duration of exposure to high dose, the timing of protocol amendments that allowed a greater proportion of patients to receive high dose, and the timing and pre-specified criteria of the futility analysis.

Bruins bepleit balans

En dat doet Bruins verdienstelijk. In zijn Kamerbrief legt de minister uit hoe de vork in de steel zit als het gaat om transparantie, intellectueel eigendom en andere stimuleringsmaatregelen, marktmacht, maximumprijzen en Europese samenwerking. De inzet van zijn beleid: innovatie stimuleren en belonen, misbruik gericht aanpakken.

Wetenschappelijk bewijs voor aanwezigheid TR4 Panama ziekte banaan in Colombia

Een internationaal team van onderzoekers heeft bewezen dat de gevreesde TR4-vorm van de schimmel die de Fusarium-verwelkingsziekte veroorzaakt die ook wel bekend staat als Panama disease, aanwezig is in Colombia. Het bewijs voor de aanwezigheid van de TR4 Fusarium odoratissimum schimmel is gepubliceerd in het tijdschrift Plant Disease.

De wetenschappelijke publicatie is de officiële bevestiging dat de gevreesde TR4-ziekteverwekker Zuid-Amerika heeft bereikt. Dat is een harde klap voor de regio, omdat de teelt en handel in bananen voor Zuid-Amerika van groot economisch en maatschappelijk belang zijn. Te meer omdat overheden en bedrijven er alles aan hebben gedaan om te voorkomen dat de ziekte zich vanuit Azië naar Zuid-Amerika zou verspreiden.

In juni 2019 werden verdachte ziektesymptomen gevonden op twee Colombiaanse bananen plantages in het district Guajira in het noordoosten van Colombia. Na deze waarnemingen ondernam de Colombiaanse overheid direct actie door de verdachte boerderijen te laten onderzoeken door het Colombiaanse instituut voor de landbouw, het ICA.

Het ICA schakelde daarbij de hulp in van Fernando Garcia-Bastidas, bananenveredelaar en Fusarium-expert bij het plantenonderzoekbedrijf KeyGene in Wageningen. Garcia reisde af naar Colombia, waar hij monsters verzamelde en de eerste tests coördineerde.

Bij dat eerste onderzoek in Colombia werd het verzamelde DNA-materiaal met verschillende technieken onderzocht. De resultaten lieten zien dat het inderdaad zeer waarschijnlijk was dat de twee verdachte plantages besmet zouden zijn met TR4.

Voor een sluitend bewijs moest in Nederland aanvullend onderzoek worden uitgevoerd bij KeyGene, Utrecht Universiteit en Wageningen University & Research. Bij het onderzoek, dat ook gecoördineerd werd door Garcia, moesten aanvullende DNA-testen worden uitgevoerd. Daarnaast moesten ook biologische toetsen worden uitgevoerd waarbij Cavendish-planten, die gevoelig zijn voor de TR4-schimmel, werden geïnfecteerd met de schimmelmonsters uit Colombia.

Het DNA-onderzoek toonde aan dat de in Colombia aangetroffen schimmel inderdaad behoort tot schimmelsoort die eerder beschreven is als Fusarium odoratissimum TR4.

In de biologische testen bleek dat alle onderzochte schimmelstammen in staat waren om TR4-gevoelige Cavendish-planten ziek te maken. De planten vertoonden van buiten én in het weefsel de typische symptomen van TR4-aantasting. Uit het zieke weefsel werd daarna schimmel-DNA geïsoleerd waarmee bevestigd werd dat de ziektesymptomen werden veroorzaakt door de schimmelstam waarmee de planten geïnfecteerd waren.

Daarmee was er sluitend bewijs dat de gevreesde TR4 inderdaad in Colombia voorkomt.

Teamwork

Aan het onderzoek en de publicatie werden vanuit Nederland bijdragen geleverd door Universiteit Utrecht (Michael Seidl & Tom Schermer), Wageningen University & Research (Gert Kema & Carolina Aguilera-Galvez),  KeyGene (eerste auteur Fernando Garcia-Bastidas, Alexander Wittenberg, Rene Hofstede, & Anker Sørensen) en ClearDetections (Marta Santos-Paiva).

Vanuit Colombia bestond het team uit onderzoekers van het ICA, het Instituto Colombiano Agropecuario (Catalina Quintero-Vargas, Mariluz Ayala-Vasquez & Anna Maria Noguera).

De auteurs van de publicatie konden, door regels vanuit het tijdschrift, in hun publicatie geen dankwoord opnemen. Het team is echter zeer erkentelijk voor de waardevolle bijdragen van de overheids-officials, de experts van het Colombian Agriculture and livestock Institute, van het ICA-hoofdkantoor en het team van het diagnostiek-laboratorium in Colombia, in het bijzonder Anamaría García, Walther Turizo, Emilio Arévalo, Jorge Palacino, Jaime Cárdenas, Alejandra Castro, Oscar Dix, Carlos Andrés Palacio en Deyanira Barrero. Cor Schoen en Yvonne Griekspoor van Wageningen University and Research hebben een actieve rol gespeeld bij het sample analyse traject.

De dreiging van TR4

TR4 is een schimmel die de dodelijke Fusarium-verwelkingsziekte veroorzaakt bij een groot aantal soorten bananenplanten. In het bijzonder planten van het bananen-ras Cavendish, dat de internationale bananenhandel domineert, zijn heel gevoelig voor TR4. TR4 is een bodemschimmel. Als de bodem eenmaal besmet is geraakt, kan een bananenteler meer dan twintig jaar geen Cavendish-bananen of andere TR4-gevoelige bananen telen. Dat is de reden dat er wereldwijd maatregelen zijn genomen om de verspreiding van TR4 te voorkomen. TR4 heeft de afgelopen twintig jaar in met name Azië en Australië talloze plantages vernield en onbruikbaar gemaakt.

TR4 is nu voor het eerst aangetoond in Zuid-Amerika. Dat is een regio die heel belangrijk is voor de internationale bananenhandel. Vanwege het grote economische en maatschappelijke belang van de bananenteelt heeft de Colombiaanse overheid op 8 augustus 2019 een nationale noodtoestand afgekondigd, toen bleek dat de laboratoriumresultaten bevestigden dat de verdachte bananenplantages met TR4 waren besmet. Het ICA, het Colombiaanse landbouw instituut, heeft becijferd dat in Colombia zo’n 175 hectares met TR4 besmet is.  

Toekomst-perspectief

Op de hele wereld wordt gewerkt aan het oplossen van de problemen met TR4. In Nederland werken KeyGene en Wageningen University & Research samen met bedrijven en instituten onder andere aan de ontwikkeling van TR4-resistente bananenrassen, grotere diversiteit in banaanrassen en beheers-strategieën.

Clémence Ross-van Dorp wordt ambassadeur sector Life, Sciences & Health

Vanaf 1 januari 2020 is Clémence Ross-van Dorp, voormalig staatsecretaris van VWS, ambassadeur voor het Actieprogramma ‘Nieuwe kansen voor Topsector Life Sciences & Health’ (LSH). Zij doet dat namens minister Bruno Bruins van Medische Zorg en Sport en staatssecretaris Mona Keijzer van Economische Zaken en Klimaat.

De aanleiding voor het opzetten van het Actieprogramma is de komst van het Europees Geneesmiddelen Agentschap (EMA) naar Nederland begin 2019. Als ambassadeur zal Ross-van Dorp zich inzetten om de economische en maatschappelijke kansen die de komst van de EMA biedt optimaal te verzilveren en zo de Nederlandse LSH-sector te versterken. Dit najaar zal Ross-van Dorp starten met voorbereidende activiteiten op haar rol als ambassadeur: zo zal ze alvast kennis gaan maken met de relevante stakeholders.

Ross-van Dorp is voorzitter van de Long Alliantie Nederland. Ook is ze bestuurder bij de Stichting Agora en vervult zij diverse toezichthoudende functies, waaronder bij de Universiteit Twente en het Prinses Máxima Centrum voor kinderoncologie. Ross-Van Dorp was van 2002 tot 2007 staatssecretaris van het ministerie van Volksgezondheid, Welzijn en Sport.

Pepscan doubles GMP production capacity to facilitate clients’ growth

Pepscan’s GMP production capacity has doubled. Hans de Backer, CEO of Pepscan, explains the new capacity additions. “Earlier this year, we started producing GMP peptides for clinical studies phases 1 and 2, which made it possible to provide peptides for purposes beyond R&D: directly for the benefit of patients. Our new production line, consisting of a new synthesizer, a preparative HPLC and lyophilizer in our cleanroom, and the strengthened QMS and process documentation that were put in place for this, have proven to facilitate the efficient execution of the process. Using this proven concept, we have now doubled the complete production line.” Current production includes both therapeutic peptide projects as well as personalized peptide medicine projects (e.g. neoantigen vaccines).

“Given our role in the field of personalized medicine, short and reliable delivery times are crucial,” adds Hanne Kho, Chief Operations Officer of Pepscan. “This demands a set of rigorous processes and a certain level of flexibility that we, together with outstanding product quality, are able to deliver. I’m very proud that the recent GMP customer audits that were conducted have confirmed this.”

ISA Pharmaceuticals receives EUR 20m loan from European Investment Bank

The European Investment Bank and ISA Pharmaceuticals have signed a EUR 20 million loan agreement to support the development of ISA’s therapy for cancers caused by the Human Papillomavirus type 16 (HPV16). The EIB’s financing is supported under the “Infectious Diseases Finance Facility” (IDFF) of the InnovFin – EU-finance for innovators programme, which is financed from the EU’s research and innovation programme Horizon 2020.

Persistent HPV16 infection can cause cervical, head & neck and anogenital cancers. These cancers can be severe and life-threatening, with low overall survival rates of advanced stages. Cervical cancer is the 2nd most common cancer in women aged 15 to 44 years in Europe, with HPV16 causing over 35,000 new cases and 15,000 deaths each year. The virus is also a major cause of oropharyngeal cancer with over 25,000 new cases and 12,000 deaths each year in Europe alone1.

ISA Pharmaceuticals has developed a novel therapy, ISA101b, which targets HPV16-positive cancers through the specific activation of the patient’s own immune system. The treatment has proven to be safe with no serious side effects, and has demonstrated substantial benefit for patients, when used in addition to current treatment in advanced stages of the head & neck cancer and cervical cancer. ISA101b represents a unique and innovative treatment modality. It is designed to fight HPV16 driven premalignant conditions and early stage cancers as monotherapy, and metastatic malignancies when given together with standard of care.

“ISA’s lead product is at a very exciting final stage of development. This loan from the EIB will help accelerate the programme all the way to our ultimate goal of making ISA101b available to all patients suffering from HPV16-induced cancers,” said Gerben Moolhuizen, CEO of ISA Pharmaceuticals.

After financing projects for research into the Zika virus and Ebola vaccines, this human papillomavirus project is one that we gladly participate in. The EIB has significantly increased its support for projects related to innovation in the health sector recently, and we see the ISA project as a very promising one with significant potential benefits for many patients.” said Ambroise Fayolle, Vice-President of EIB.

Through the support of IDFF, the EIB can provide stable long-term funding with a flexible repayment and interest structure, limiting the cash outflow from the company and thereby enabling it to focus on investing in innovation and growth.

Carlos Moedas, European Commissioner for Research, Science and Innovation, said: “With more people living longer, cancer rates are increasing everywhere. Europe has 9% of the world’s population but a 23% share of the global cancer burden. This is why we support the development of new drugs to treat life-threatening diseases such as cancer through the EU research and innovation programme, Horizon 2020.”

Van volume- naar waardegedreven zorg

De NZa spoort ziekenhuizen en verzekeraars opnieuw aan om de gezondheidswinst voor de patiënt centraal te stellen in de contractering van de medisch-specialistische zorg. Zij herhaalt daarmee de boodschap uit haar vorig jaar verschenen advies “belonen van zorg die waarde toevoegt”. In dit rapport pleit de NZa voor een transitie van volume- naar waardegedreven zorg. Een mooie ontwikkeling, HollandBIO is blij is dat de NZa voor de patiënt op de bres springt. Wij geloven dat deze vlieger ook elders in de zorg opgaat en maken daar sinds jaar en dag werk van. In ons ideale zorgsysteem gaan gezondheidswinst, innovatie en betaalbaarheid namelijk hand in hand!
