Harvard and MIT Experts Develop New Gene Editing Technology

Experts from Broad Institute of Massachusetts Institute of Technology (MIT) and Harvard University developed a new gene editing approach by putting together two of the most vital proteins in molecular biology, CRISPR-Cas9 and a reverse transcriptase, into one system. The new system called prime editing is capable of editing mammalian cells in a precise, efficient, and highly versatile fashion. The study was published in Nature.

The main difference of prime editing from previous genome editing systems is that it uses RNA to direct the insertion of new DNA sequences in human cells. The new system involves combining Cas9 with reverse transcriptase. The molecular complex uses one strand of the target DNA site to “prime,” or initiate, the direct writing of edited genetic information into the genome. The system also uses a a new type of engineered guide RNA (pegRNA), which directs the prime editor to its target site where a modified Cas9 cuts one strand of the DNA.

Now online: HollandBIO Highlights English edition

Biotech volop in de aandacht tijdens Dutch Design Week

Afgelopen week vond de jaarlijkse editie van de Dutch Design Week plaats. Net als in eerdere edities, kwam biotechnologie als rode draad terug in verschillende toepassingen, exposities en discussies. Dit leverde interessante perspectieven en ideeën op. Want waar biotechnologie van oudsher in hokjes als geneesmiddelen en planten en industriële toepassingen valt, komen tijdens de Dutch Design Week heel andere benaderingen naar voren. Wat te denken van de bioplastics uit koeienmest waar kunstenaar en ondernemer Jalila Essaïdi aan werkt? Of de ‘levende’ huizen van de toekomst die architect Bob Hendrikx wil ontwikkelen met hulp van biotech.

MSD breidt flink uit in Haarlem

Bij de MSD-locatie in Haarlem ging afgelopen week de eerste paal de grond in voor een gloednieuw koelmagazijn vlak achter het huidige exemplaar. Ook start binnenkort de bouw van een laboratorium en een omvangrijke uitbreiding van de huidige fabrieksruimte. Met de uitbreidingen speelt MSD in op de snelgroeiende vraag naar vaccins, waarvoor het bedrijf in de komende 3 tot 5 jaar een verdubbeling van de productie verwacht.

Daarnaast bereidt MSD zich voor op een mogelijke harde Brexit, wat zou betekenen dat in het Verenigd Koninkrijk verpakte medicijnen voor de Europese markt onder andere in Haarlem opnieuw getest moeten worden. Om de medicijnen adequaat te bewaren, is een nieuw koelmagazijn noodzakelijk. MSD is met 1000 werknemers in Haarlem al een van de grootste werkgevers in de regio, maar versterkt deze positie met deze uitbreiding nog verder.

Cato Research and SMS-oncology merge

Cato Research has merged with Specialized Medical Services-oncology BV (SMS-oncology) to bolster its portfolio of global solutions dedicated to supporting life science organizations with advancing drug development. The combined company will transition to the new name, CATO SMS, in the coming year.

By joining forces, CATO and SMS-oncology expand their presence across Europe and North America, deepen their oncology and regulatory expertise, and broaden their suite of services. Blokhuis: Pasgeboren baby’s testen op SMA met hielprik

Staatssecretaris Paul Blokhuis wil dat de ziekte spinale musculaire atrofie (SMA) wordt opgenomen in de hielprikscreening bij pasgeboren baby’s. De staatssecretaris schrijft aan de Tweede Kamer dat hij het RIVM vraagt om de nodige zaken uit te zoeken en voor te bereiden. Hiermee volgt Blokhuis het advies op van de Gezondheidsraad.

SMA is een ernstige spierziekte die verlamming en overlijden tot gevolg kan hebben. De ziekte openbaart zich op jonge leeftijd en wordt nu pas vastgesteld als er symptomen optreden. De schade die dan is aangericht is onherstelbaar. Door de ziekte toe te voegen aan de testen hielprikscreening, kan de ziekte worden vastgesteld en behandeld vóórdat het zenuwstelsel onomkeerbaar beschadigd raakt. Er is sinds kort een medicijn opgenomen in het basispakket dat deze schade kan beperken of zelfs helemaal voorkomen als het wordt toegediend voordat er symptomen optreden. Naar verwachting kan met de hielprikscreening SMA jaarlijks bij ongeveer vijftien tot twintig kinderen worden vastgesteld.

Blokhuis: “Door SMA op te nemen in de screening kan de ziekte al heel vroeg in het leven van kinderen worden vastgesteld én worden behandeld. We kunnen zo het leed van kinderen met deze ziekte verminderen of zelfs helemaal voorkomen. Ik heb het RIVM verzocht om uit te zoeken wat er allemaal nodig is om SMA op te nemen in de screening en om hiervoor alvast voorbereidingen te treffen.”

RIVM

Om de procedure in gang te zetten vraagt Blokhuis het RIVM om een zogenoemde uitvoeringstoets uit te voeren. Daarin beschrijft het RIVM wat er moet gebeuren om SMA te implementeren, hoe lang daarvoor nodig is en hoeveel het kost. Daarbij zal het RIVM een testmethode uitzoeken en valideren. Ook stelt het RIVM een protocol vast hoe de zorg eruitziet als een patiënt wordt gediagnosticeerd.

Hielprikscreening

In de eerste week na de geboorte wordt de hielprik afgenomen. Bij de huidige screening wordt het bloed getest op 22 ernstige en zeldzame ziektes. Sinds afgelopen oktober zijn er drie ziektes toegevoegd en de komende jaren komen daar naast SMA nog negen bij.

Naar verwachting zal het RIVM Blokhuis in het najaar 2020 op de hoogte brengen van de uitkomsten van de uitvoeringstoets. Dan zal er ook meer duidelijkheid zijn over wanneer SMA in de hielprikscreening zal worden opgenomen.

How Seaweed Blends Enhance the Animal Microbiome

Ocean Harvest Technology produces seaweed-based functional feed ingredients for animals. Together with BaseClear, the company is now investigating the effects of its seaweed blend on the animal microbiome and health. “On the market you hear a lot of claims about new gut health ingredients, but few of these claims are backed by clear evidence. At Ocean Harvest, we work with BaseClear to create such data so that when we say we have a strong prebiotic, we can prove it.”

Ocean Harvest Technology develops and manufactures animal feed ingredients based on seaweed. Its product range consist of seaweed blends for livestock and companion animals, which are based on a mixture of different species of brown, green and red macroalgae. OceanFeed™ blends contain prebiotic polysaccharides and other natural bioactive components that enhance gut microflora and not only support a positive immune response but also a healthier gut in general.

FOCUS ON ANIMAL MICROBIOME

There is an increasing interest in the status of the gut microflora of animals and the effect of feed on this indicator of gut health. Jason Sands, product portfolio manager at Ocean Harvest Technology in the UK, explains: “For a long time, farmers relied on antibiotics and other chemical additives to ensure animal health. However, the overuse of antibiotics has led to resistance and regulators are now banning them in an increasing number of countries. Farmers are therefore in need of more sustainable solutions to improve the health of their animals. The analysis of the microbiome can play an important role in that, since an animal’s health is strongly affected by the status of its microbiome.”

Derek Butler, commercial director at BaseClear, confirms this. “We are observing an increasing demand for the analysis of the microbiome of animals. While a few years ago, this type of analysis was mainly requested for model animals, now we see a growing interest in the analysis of the microbiome of agricultural animals. Sometimes the questions are related to antibiotic resistance and residual antibiotics, while in other cases it is more related to feed conversion rates.”

Graham Ellis, CEO of Ocean Harvest Technology, expects that in the near term, the analysis and knowledge of the animal microbiome will prevail over the analysis of the human microbiome, although currently the analysis of the human microbiome is technically leading. “We have much better control over the diet and living conditions of animals, while humans have different diets and different stress conditions. It is therefore significantly easier to obtain statistical outcomes from the analysis of the animal microbiome versus that of humans.”

SCIENTIFIC BASIS FOR HEALTH CLAIMS

Ocean Harvest Technology started a collaboration with BaseClear in 2018, since they didn’t want to make claims about the prebiotic effects of their animal feed ingredients without having scientific backing. “On the market you hear a lot of claims about new gut health ingredients, but few of these claims are backed by clear evidence. At Ocean Harvest, we work with BaseClear to create such data so that when we say we have a strong prebiotic, we can prove it.” says Ellis. The company has already demonstrated the effect of its seaweed blends on animal performance, but now they also want to show the underlying effect on the animal microbiome.

Within this project, BaseClear started to analyse the microbiome of pigs and poultry using 16S profiling. For this, Ocean Harvest Technology delivered samples of animals that were fed with normal feed and samples of animals that were fed with seaweed-containing feed. Sands elaborates: “There is more and more public information about how a healthy microbiome should look, and there is also growing evidence about how microbiome profiles correlate with the overall performance of animals. That is what we want to use for our research. We want to correlate microbiome profiles to the overall health and performance of animals, such as their feed efficiency, and see how this is affected by our seaweed blends.”

“In principle, we could even go one step further and do a deeper type of characterisation based on metagenomics and transcriptomics. That would provide even more information about what the bacteria in the gut are doing,” Butler elaborates.

MICROBIOME ANALYSIS AS A TOOL FOR CUSTOMISATION

When talking about the future vision, the ideal picture would be to use real-time animal microbiome analysis as a tool to offer farmers a customised or variable animal feed blend based on the analysis of their own animals. “This is an exciting idea. But it is therefore necessary to reduce the turnaround times and prices of the microbiome analysis. The state it is in now is fine for research and development, but not for the use as a tool for our clients,” Ellis says. Butler expects that this will indeed be possible in the future. “In fact, the technology for this is already available, namely the Oxford Nanopore sequencing technology, which we also have available in our lab,” he says.

SAMPLING AT THE FARM

“This collaboration project wouldn’t have been possible without the readymade RNA sample stabilisation tubes that clients of BaseClear can use to send their samples to the BaseClear laboratory”, says Ellis. “Many companies and institutes request clients to send the samples on dry ice or liquid nitrogen, which wasn’t really an option for us. You can’t go to a farmer and ask if he doesn’t mind taking a sample and storing it at minus eighty degrees! It wasn’t widely known that there are alternatives for that. We were very happy to discover that BaseClear were willing to accept sample tubes that can be transported at ambient temperature, which enabled us to do the research in the way that we wanted.”

SUMMARY

Ellis concludes: “At Ocean Harvest, we’re very proud to be able to share these type of analyses with our customers and those intending to trial our products. Invariably, it’s the first time that they’ve seen data this clear and conclusive. We’re extremely happy that our collaboration with BaseClear is helping farmers make a confident decision to reformulate away from problematic ingredients like antibiotics, and to use natural options which are fundamentally enhancing the animal’s health and performance.”

FDA wil status quo blijven doorbreken rond regulatoire wetenschap

De Food and Drug Administration (FDA) wil ook in de toekomst toonaangevend blijven op het gebied van regulatoire wetenschap. De Amerikaanse geneesmiddelenautoriteit kondigt daartoe de nodige veranderingen in organisatie en werkwijze aan. Het lezen van de rationale achter die ambitie was een feest der herkenning voor HollandBIO. De FDA erkent dat de ‘gouden standaard’ continu aan verandering onderhevig is, en concludeert dat ook zij daarom de regulatoire status quo moet blijven doorbreken. Waar kennen we dat motto ook alweer van?

Hoewel de aankondiging van de FDA, getiteld “Reorganization of the Office of New Drugs with Corresponding Changes to the Office of Translational Sciences and the Office of Pharmaceutical Quality,” niet heel spannend klinkt, betekent deze weldegelijk een doorbraak, zo stelt Peter J. Pitts in een column in Statnews. En Pitts kan het weten: hij is onder meer president van het Center for Medicine in the Public Interest en was associate commissioner bij de FDA. Naast een vergroting van capaciteit voor de beoordeling van nieuwe geneesmiddelen en klinisch onderzoek, toont de FDA met de plannen dat zij echt leiderschap wil tonen. Niet omdat de FDA die met het oog op haar rol sowieso heeft, maar op basis van haar excellente regulatoire expertise. Hear hear!

VWS publiceert vernieuwde Regeling Voorwaardelijke Toelating

Op 22 oktober stuurde minister Bruins (Medische Zorg) het langverwachte beleidskader Voorwaardelijke Toelating (VT) geneesmiddelen naar de Tweede Kamer. Met de nieuwe regeling VT wil de minister de toegang tot nieuwe geneesmiddelen verbeteren voor patiënten met ernstige en vaak zeldzame aandoeningen. Een streven dat op de volle steun van HollandBIO kan rekenen. En ook over de uitwerking van de regeling, die per direct ingaat, zijn we voorzichtig enthousiast. Maar, as always, the proof of the pudding is in the eating.

Weesgeneesmiddelen zijn de innovatieve kraamkamer van de life sciences sector. Baanbrekende nieuwe behandelingen bewijzen zich doorgaans als eerste in het zeldzame ziektedomein, voor ze breder toegepast worden. Dankzij gerichte stimuleringsmaatregelen en onze toenemende kennis van de biologische processen die ten grondslag liggen aan ziekte, is de life sciences sector steeds beter in staat geneesmiddelen en therapieën te ontwikkelen die aansluiten op de individuele behandelbehoefte van de patiënt.

De ontwikkeling van weesgeneesmiddelen stelt alle facetten van het klassieke traject van geneesmiddelenontwikkeling, beoordeling en toegang op de proef. Gouden standaarden, ontwikkeld voor blockbusters, passen niet meer. Ze zijn onhaalbaar of zelfs onethisch. Personalised medicine vergt meer dan alleen de ontwikkeling van geneesmiddelen op maat. Ook het systeem van beoordeling en vergoeding moet mee innoveren.

Die aansluiting laat volop ruimte voor verbetering. Waar het Europees Medicijn Agentschap (EMA) diverse routes heeft ontwikkeld voor een beoordeling op maat, blijft het nationale traject voor toegang en financiering van geneesmiddelen vooralsnog vasthouden aan een gedateerd one size fits all beoordelingskader. Geregistreerde weesgeneesmiddelen, al dan niet conditional of exceptional, lopen daardoor stuk op de beoordeling van het Zorginstituut. Het resultaat is schrijnend: voor patiënten met een zeldzame ziekte is eindelijk een geregistreerd geneesmiddel op de markt, maar de toegang loopt enorme vertraging op of strandt in het zicht van de haven.  

Als één van de manieren om dit probleem aan te pakken, greep HollandBIO de herinrichting Regeling voorwaardelijke toelating tot het basispakket, die het ministerie van VWS in februari 2017 aankondigde, met beide handen aan. Voor, tijdens en na de consultatie pleitten we voor een innovatieve en flexibele regeling die beter zou aansluiten bij EMA registratie, maar die vooral ook het onderliggende methodologische probleem zou erkennen en aanpakken. Onder meer door ruimte te laten voor maatwerk, in studie ontwerp, methodologie en beoordelingskaders.

We zijn blij dat het nu gepubliceerde beleidskader en de uitvoeringstoets het door de sector aangekaarte methodologische probleem onderschrijft en de flexibiliteit belooft die de nieuwe generatie geneesmiddelen nodig heeft. Wel stelt het ministerie fikse voorwaarden in ruil voor voorwaardelijke toelating: over prijsstelling, transparantie en openbaarheid van data. Alleen de praktijk kan uitwijzen of de voordelen opwegen tegen de nadelen.

De nieuwe regeling VT lijkt in ieder geval een oplossing voor die geneesmiddelen, die nu echt stranden in het rigide nationale systeem voor toelating. Maar hopelijk groeit hij uit tot meer. Als het aan ons ligt, is de vernieuwde regeling VT de opmaat naar een adaptief ecosysteem, waarin registratie naadloos aansluit op toegang en vergoeding. Daarmee bieden we de beste zorg aan patiënten met een zeldzame aandoening, én zet Nederland zichzelf op de kaart als innovatieve proeftuin voor weesgeneesmiddelen, en de voorhoede van de transitie naar personalised medicine die zij vertegenwoordigen.

Syngenta pakt klimaatverandering aan

Syngenta heeft vandaag aangekondigd dat het in de loop van de komende vijf jaar 2 miljard dollar zal uittrekken om boeren te helpen zich voor te bereiden op de toenemende dreiging van klimaatverandering en deze het hoofd te bieden.

De investering ondersteunt een nieuw duurzaamheidsdoel van Syngenta om jaarlijks ten minste twee technologische doorbraken op de markt te brengen, de bijdrage van de landbouw aan klimaatverandering te verminderen, het mitigerende vermogen van de sector in te zetten en te helpen het voedselsysteem binnen het draagvermogen van de planeet te houden.

Erik Fyrwald, Chief Executive Officer bij Syngenta, kondigde bovendien aan dat de investering in onderzoek en ontwikkeling voor duurzame landbouw gepaard zal gaan met het streven om tegen 2030 de CO2-intensiteit van de operationele activiteiten van het bedrijf met ten minste 50 % te verminderen en daarmee de ambitieuze doelen van het Akkoord van Parijs inzake klimaatverandering te ondersteunen. De toezegging van Syngenta is gevalideerd en goedgekeurd door het Science Based Targets initiative (SBTi).

“De landbouw bevindt zich nu in de frontlinie van de wereldwijde inspanningen tegen klimaatverandering”, aldus de heer Fyrwald. “ Syngenta legt zich toe op versnelde innovatie om betere en steeds veiligere oplossingen te vinden om de gemeenschappelijke uitdaging van klimaatverandering en verlies van biodiversiteit aan te gaan.

En we laten het niet bij woorden: Syngenta onderneemt concrete actie om boeren te helpen klimaatverandering tegen te gaan en de bijdrage van de sector aan de wereldwijde emissies van broeikasgassen te verminderen.”

De 2 miljard dollar zal worden aangewend voor programma’s met duidelijk gedifferentieerde voordelen of baanbrekende technologieën die een sprong voorwaarts mogelijk zullen maken in de duurzaamheid van de landbouw, zoals landgebruik, bodemgezondheid en geïntegreerde plagenbestrijding.

Dankzij een meerjarige samenwerking met The Nature Conservancy ontwikkelt Syngenta strategieën om nieuwe innovaties en technologieën aan te wijzen ten gunste van boeren en het milieu. De samenwerking vindt zijn oorsprong in inspanningen ter verbetering van bodemgezondheid, efficiëntie van hulpbronnen en habitatbescherming in belangrijke landbouwgebieden wereldwijd.

Sally Jewell, CEO van The Nature Conservancy: “Om natuurbehoud op schaal te bewerkstelligen is vastberaden actie van de private sector nodig. Nu ondernemingen de risico’s van klimaatverandering en de voordelen van duurzaamheid steeds meer onderkennen, zijn wij blij met deze gelegenheid om met onze wetenschappelijke kennis en expertise bij te dragen aan het transformeren van de werkwijzen in de sector. De investering van Syngenta in innovatie is een belangrijke stap in de richting van een toekomst waarin de natuur en de mensheid samen kunnen gedijen.”

Cynthia Cummis, directeur Private Sector Climate Mitigation bij World Resources Institute, een van de partners bij Science Based Targets initiative: “Het is te prijzen dat Syngenta haar emissiereductiedoelstellingen door Science Based Targets initiative heeft laten valideren. Leiderschap van de kant van de agro-industrie is onmisbaar in de strijd tegen klimaatverandering. Door deze doelen te stellen slaat Syngenta het pad in naar toekomstbestendige groei.”

De doelstellingen maken deel uit van de eerder dit jaar door Syngenta gelanceerde toezegging “Accelerating Innovation” om de toegenomen uitdagingen aan te gaan waar boeren mee te kampen hebben als gevolg van klimaatverandering, bodemerosie en verlies van biodiversiteit. Er zal jaarlijks verslag worden gedaan van de voortgang ten opzichte van deze doelstellingen. Op deze voortgang vindt een onafhankelijke audit plaats. De aankondiging is hoofdzakelijk gebaseerd op 150 luistersessies die wereldwijd zijn gehouden en op basis waarvan de directie van het bedrijf prioriteitsgebieden voor investering heeft aangewezen.
