Elke andere uitdaging is oplosbaar.

Crisp & malty. Your first CRISPR beer

This is probably the first product branded as CRISPR-edited, how cool is it? The science comes from Berkeley Yeast. The cans are sold by Temescal Brewing and the claim is “Crisp, Malty, Helles Lager”.

Further details about CRISPR 5,2%: “A floor-malted bohemian pilsner is a process-driven beer with a simple recipe of just floor-malted bohemian pilsner malt, Saaz hops, lager yeast, and water. We carefully step mash the malt to get a deeper, breadier flavor, while keeping it crisp and light-bodied. A light touch of Saaz hops balances the beer with some spicy and floral notes. A longer boil brings out more honey notes from the malt. It’s fermented cold with a CRISPR-modified Lager yeast, designed to prevent the production of diacetyl. We naturally carbonated this beer for great mouthfeel and head retention and patiently lagered it for 3 weeks, allowing the beer to mellow and mature”.

Curious to know why diacetyl-free beer is a good idea? Here is the answer: “Diacetyl is a common off-flavor in beer. At high concentrations it resembles artificial butter and butterscotch. At lower concentrations it presents as a buttery slickness on the tongue or roof of the mouth. Even at low concentrations, it distracts from an otherwise amazing IPA.”

The Berkeley researchers have more to offer. See their beer strains Wildflower, Melon Drop, Galactic, and Berry Jammer. And check A CRISPR Shade of Blonde, with subtle floral and fruit notes from one of these special strains.

Moderna’s COVID-19 Vaccine Candidate Meets its Primary Efficacy Endpoint in the First Interim Analysis of the Phase 3 COVE Study

Moderna, Inc. (Nasdaq: MRNA), a biotechnology company pioneering messenger RNA (mRNA) therapeutics and vaccines to create a new generation of transformative medicines for patients, announced that the independent, NIH-appointed Data Safety Monitoring Board (DSMB) for the Phase 3 study of mRNA-1273, its vaccine candidate against COVID-19, has informed Moderna that the trial has met the statistical criteria pre-specified in the study protocol for efficacy, with a vaccine efficacy of 94.5%. This study, known as the COVE study, enrolled more than 30,000 participants in the U.S. and is being conducted in collaboration with the National Institute of Allergy and Infectious Diseases (NIAID), part of the National Institutes of Health (NIH), and the Biomedical Advanced Research and Development Authority (BARDA), part of the Office of the Assistant Secretary for Preparedness and Response at the U.S. Department of Health and Human Services.

The primary endpoint of the Phase 3 COVE study is based on the analysis of COVID-19 cases confirmed and adjudicated starting two weeks following the second dose of vaccine. This first interim analysis was based on 95 cases, of which 90 cases of COVID-19 were observed in the placebo group versus 5 cases observed in the mRNA-1273 group, resulting in a point estimate of vaccine efficacy of 94.5% (p <0.0001).

A secondary endpoint analyzed severe cases of COVID-19 and included 11 severe cases (as defined in the study protocol) in this first interim analysis. All 11 cases occurred in the placebo group and none in the mRNA-1273 vaccinated group.

The 95 COVID-19 cases included 15 older adults (ages 65+) and 20 participants identifying as being from diverse communities (including 12 Hispanic or LatinX, 4 Black or African Americans, 3 Asian Americans and 1 multiracial).

The interim analysis included a concurrent review of the available Phase 3 COVE study safety data by the DSMB, which did not report any significant safety concerns. A review of solicited adverse events indicated that the vaccine was generally well tolerated. The majority of adverse events were mild or moderate in severity. Grade 3 (severe) events greater than or equal to 2% in frequency after the first dose included injection site pain (2.7%), and after the second dose included fatigue (9.7%), myalgia (8.9%), arthralgia (5.2%), headache (4.5%), pain (4.1%) and erythema/redness at the injection site (2.0%). These solicited adverse events were generally short-lived. These data are subject to change based on ongoing analysis of further Phase 3 COVE study data and final analysis.

Preliminary analysis suggests a broadly consistent safety and efficacy profile across all evaluated subgroups.

As more cases accrue leading up to the final analysis, the Company expects the point estimate for vaccine efficacy may change. The Company plans to submit data from the full Phase 3 COVE study to a peer-reviewed publication.

“This is a pivotal moment in the development of our COVID-19 vaccine candidate. Since early January, we have chased this virus with the intent to protect as many people around the world as possible. All along, we have known that each day matters. This positive interim analysis from our Phase 3 study has given us the first clinical validation that our vaccine can prevent COVID-19 disease, including severe disease,” said Stéphane Bancel, Chief Executive Officer of Moderna. “This milestone is only possible because of the hard work and sacrifices of so many. I want to thank the thousands of participants in our Phase 1, Phase 2 and Phase 3 studies, and the staff at our clinical trial sites who have been on the front lines of the fight against the virus. They are an inspiration to us all. I want to thank the NIH, particularly NIAID, for their scientific leadership including through years of foundational research on potential pandemic threats at the Vaccine Research Center that led to the discovery of the best way to make Spike protein antigens that are being used in our vaccine and others’. I want to thank our partners at BARDA and Operation Warp Speed who have been instrumental to accelerating our progress to this point. Finally, I want to thank the Moderna team, our suppliers and our partners, for their tireless work across research, development and manufacturing of the vaccine. We look forward to the next milestones of submitting for an EUA in the U.S., and regulatory filings in countries around the world, while we continue to collect data on the safety and efficacy of the vaccine in the COVE study. We remain committed to and focused on doing our part to help end the COVID-19 pandemic.”

Based on these interim safety and efficacy data, Moderna intends to submit for an Emergency Use Authorization (EUA) with the U.S. Food and Drug Administration (FDA) in the coming weeks and anticipates having the EUA informed by the final safety and efficacy data (with a median duration of at least 2 months). Moderna also plans to submit applications for authorizations to global regulatory agencies.

Moderna is working with the U.S. Centers for Disease Control and Prevention (CDC), Operation Warp Speed and McKesson (NYSE: MCK), a COVID-19 vaccine distributor contracted by the U.S. government, as well as global stakeholders to be prepared for distribution of mRNA-1273, in the event that it receives an EUA and similar global authorizations. By the end of 2020, the Company expects to have approximately 20 million doses of mRNA-1273 ready to ship in the U.S. The Company remains on track to manufacture 500 million to 1 billion doses globally in 2021. On November 10, the American Medical Association (AMA) issued a Current Procedural Terminology (CPT) code to report vaccination with mRNA-1273 (code: 91301). Moderna recently announced further progress towards ensuring the distribution, storage and handling of the vaccine can be done using existing infrastructure.

  • versterkte paraatheid: met het oog op de vaststelling van plannen op nationaal niveau zal er op EU-niveau een gezondheidscrisis- en pandemieparaatheidsplan worden ontwikkeld en zullen in deze context aanbevelingen worden gedaan, in combinatie met uitgebreide, transparante kaders voor verslaglegging en controle. De opstelling van nationale plannen wordt ondersteund door het Europees Centrum voor ziektepreventie en -bestrijding en andere EU-agentschappen. De plannen worden gecontroleerd en gestresstest door de Commissie en de EU-agentschappen;
  • versterkt toezicht: op EU-niveau zal een versterkt, geïntegreerd bewakingssysteem worden opgezet, waarbij kunstmatige intelligentie en andere geavanceerde technologische middelen worden ingezet;
  • verbeterde gegevensrapportering: van de lidstaten wordt verlangd dat zij hun rapportage van indicatoren voor gezondheidszorgstelsels intensiveren (bv. beschikbaarheid van ziekenhuisbedden, capaciteit voor gespecialiseerde behandeling en intensieve zorg, aantal medisch opgeleide personeelsleden enz.);

  • epidemiologische surveillance via geïntegreerde systemen die realtime surveillance mogelijk maken;
  • paraatheids- en responsplanning, verslaglegging en controle;
  • het geven van niet-bindende aanbevelingen en opties voor risicobeheer;
  • capaciteit om de EU-taskforce voor gezondheid te mobiliseren en in te zetten om de lokale respons in de lidstaten te ondersteunen;

  • het risico op tekorten aan kritieke geneesmiddelen en medische hulpmiddelen te monitoren en te beperken;
  • wetenschappelijk advies te geven over geneesmiddelen die mogelijk de ziekten die deze crises veroorzaken, kunnen behandelen, voorkomen of helpen diagnosticeren;
  • studies te coördineren om de doeltreffendheid en veiligheid van vaccins te controleren;

AM-Pharma Announces Enrollment of the First Patient in REVIVAL Phase III Pivotal Trial in Patients with Sepsis-Associated Acute Kidney Injury

AM-Pharma B.V., an emerging leader focused on the treatment of kidney disease, sepsis and organ injury, today announced that it has enrolled the first patient in its global Phase III trial named REVIVAL (REcombinant human alkaline phosphatase SA-AKI surVIVAL trial). The REVIVAL trial will evaluate the Company’s proprietary recombinant alkaline phosphatase for the treatment of SA-AKI patients with the primary objective to confirm the reduction in mortality as demonstrated in the Company’s Phase II STOP-AKI trial[1]. Clinical trial sites across Europe, the United Kingdom and North America will enroll up to 1,600 SA-AKI patients.

“Sepsis-associated acute kidney injury is the cause of death for hundreds of thousands of people hospitalized each year. Currently, there are no treatments available outside of supportive care in an ICU,” stated Erik van den Berg, Chief Executive Officer at AM-Pharma. “Our compound represents an opportunity to reduce the high mortality rates and protect kidney function for patients with SA-AKI. Over the last 18 months, our focus has been to prepare a rigorous pivotal trial together with global AKI and sepsis experts, building on the strong Phase II STOP-AKI data and constructive interactions with the FDA and EMA. I would like to thank the AM-Pharma team, the clinical centers and the trial investigators for all their hard work and collaboration to achieve this important milestone today.”

The REVIVAL Phase III pivotal trial is a randomized, double-blind, placebo-controlled, two-arm, parallel-group, multi-center trial to evaluate the efficacy and safety of AM-Pharma’s proprietary human recombinant alkaline phosphatase for the treatment of patients with SA-AKI. The study will enroll approximately 1,400 patients with SA-AKI in the main study population. In two exploratory cohorts, up to 100 patients with moderate Chronic Kidney Disease (CDK) and up to 100 patients with COVID-19 will be enrolled. The primary aim of the study is to confirm the improvement on the primary endpoint of 28-day all-cause mortality, as observed in the Phase II STOP-AKI study. Secondary endpoints include the treatment effect on long-term Major Adverse Kidney Events (MAKE), organ support use, length of stay in the ICU and 90-day all-cause mortality. Further information on this study can be found at www.clinicaltrials.gov, NCT04411472 (REVIVAL).

“From the exciting results we have seen in the Phase II STOP-AKI trial, AM-Pharma’s drug candidate has the potential to transform the treatment landscape for patients with sepsis-associated acute kidney injury,” stated Professor Peter Pickkers, M.D., Ph.D., Chair of Experimental Intensive Care Medicine, Radboud University Medical Center, and principal investigator of the REVIVAL study. “In the Phase II STOP-AKI trial, treatment with AM-Pharma’s recombinant alkaline phosphatase resulted in a relative reduction in mortality of more than 40% and sustained improvement in renal function over the course of the 28-day study period. Based on these promising results, we are excited to be part of this pivotal study and the innovation we believe it can bring to SA-AKI treatment.”

“Sepsis is a highly prevalent, severe condition that causes long-term physical impact on patients who survive their admission to the ICU. The fact that there are currently no approved treatments available underscores the high unmet medical need for patients. We look forward to working together with AM-Pharma to potentially bring this treatment option to patients,” added John A. Kellum, M.D., Professor, Vice Chair Department of Critical Care Medicine and Director at the Center for Critical Care Nephrology at University of Pittsburgh.

For the Phase III REVIVAL trial, potentially over 100 sites across Europe, the United Kingdom and North America are actively recruiting patients with SA-AKI in the trial. Clinical site activation is progressing as planned and topline safety and futility analyses on the first 400 patients in the main study population is expected by the end of 2021. The Company expects to complete target enrollment and to announce data on the primary endpoint of 28-day all-cause mortality in 2023.

Acute Kidney Injury (AKI) involves inflammatory processes in the kidney which can lead to complete loss of renal function. Hospital‐acquired AKI affects annually around 3 million patients in Europe, the US and Japan, and is associated with mortality in roughly 700,000 patients. It occurs in 40-60% of critical care admissions. Depending on the severity and cause of renal injury, mortality ranges from 10% to as high as 60%. In the US alone, hospitals spend around $10 billion each year on managing this major medical problem.[2],[3],[4]

Sepsis is a condition that is responsible for 1 out of 3 deaths in hospitals and is defined as life-threatening organ dysfunction caused by a dysregulated host response to infection. The kidney is the most commonly affected organ, resulting in SA-AKI and significantly increasing the risk for mortality and morbidity in sepsis. No singular effective therapy to alter the progression of these devastating conditions has been approved, while the healthcare burden for sepsis in the US alone is $16.7 billion on an annual basis.[5]

AM-Pharma’s therapeutic candidate is a proprietary recombinant human Alkaline Phosphatase (AP) constructed from two naturally occurring human isoforms of the AP enzyme. The Company’s compound is highly stable and active and has a dual mechanism of action via dephosphorylation of lipopolysaccharides (LPS) and extracellular ATP. AM-Pharma has shown that treatment of patients with exogenous AP not only reduces local and systemic inflammation but also protects the kidney against further damage.

AM-Pharma’s purpose is to save and improve the lives of patients confronted with kidney disease, sepsis and organ injury. Our initial focus is sepsis-associated acute kidney injury, the cause of death for hundreds of thousands of people hospitalized each year. Our proprietary recombinant human alkaline phosphatase has the potential to become the first treatment for sepsis-associated acute kidney injury and is now in a global pivotal Phase III clinical trial. We are a dedicated team driven to bring treatment options to severely ill patients, their families and acute care professionals.

  • Additional questions of your choosing tied to strategy, finance, competitive intelligence, marketing…

It is an occasion for you to propose a project free of charges but also to know our students and our master.

As a list of projects will be sent to our students on the 6th of December, we would ask you to send us a draft of the project before the 28th of November to have enough time to adjust together the project to the requirements of the master.

For this project, we would request from your company:

  • A 4-8 PowerPoint slide presentation in English that explains what your company does and what your technology is (a short corporate presentation also works well)
    • The document needs to include specific questions that you would like the students to work on. (They will automatically prepare a technological comparison and a market analysis).
    • We can help you writing the questions so that there are no misunderstandings about the deliverables
    • It also needs to include the contact information of the persons that will interact with the students
  • Be available to answer questions about your company, or about the industry that you’re working in.
  • Give some sort of basic feedback about what you thought of the work the students did for you.

We can send you examples of questions asked to the students last year.
