EFSA: Existing guidance appropriate for gene editing assessment

Genome editing does not pose any additional hazards compared to conventional breeding or other genetic modification methods, according to the European Food Safety Agency (EFSA) in a scientific opinion requested by the European Commission. Therefore, the existing guidance documents for risk assessment are sufficient but only partially applicable to plants developed using genome editing, as requirements relating to foreign DNA are not relevant.

EFSA’s latest scientific opinion, published on Wednesday 24 November, focused on plants produced using different genome editing techniques. These techniques modify a specific region of the genome but, unlike traditional genetic modification (GM), do not introduce foreign DNA.

The opinion was requested by the European Commission, who asked EFSA to assess whether its guidelines for the risk assessment of GM plants can be used for the risk assessment of plants produced using these techniques.

Experts concluded that the existing guidance is applicable for the evaluation of the three new techniques. In fact, EFSA concluded that fewer datasets may be needed for the risk assessment compared to other GM techniques given the absence of new DNA.

Daarvan is ruim 900.000 euro Europese subsidie (EFRO).

TargED closes €1.35mln seed investment and partners with UMC Utrecht for the development of Microlyse

TargED Biopharmaceuticals, a biotech spin-off of the University Medical Center Utrecht founded by Associate prof. Coen Maas, PhD, Steven de Maat, PhD, Marc van Moorsel and Kristof Vercruysse, raised EUR 1.35 million to develop a unique biotechnological drug for rapid degradation of blood clots that obstruct the vasculature. The funds include a seed investment from FIRST fund, Curie Capital and Utrecht Health Seed Fund as well as a public-private-partnership with UMC Utrecht, supported by additional funding from Health Holland.

Every year, approximately 1.3 million Europeans and 800.000 US citizens have a first stroke. Stroke is the second most common cause of death and a major cause of disabilityin developed economies. The majority of stroke events are caused by an occlusion of a blood vessel(thrombosis). TargED’s Microlyse, a new proprietary clot-busting compound, binds to a protein that is present in all forms of thrombosis, ranging from those that obstruct the largest vessels to the smallest vessels in the body (microvasculature). Due to its unique mechanism of action, Microlysecan stimulate a natural pathway that dissolves these blood clots. This therapeutic approach can be used torapidlyeliminateocclusive blood clots and has the potential to outperform current stroke caretherapeutics.Next to acute ischemic stroke, thrombosis is seen in a wide range of cardiovascular conditions for which TargED expects that Microlyse is also applicable, such as the life-threateningdisease TTP (thrombotic thrombocytopenic purpura).

Kristof Vercruysse, interim CEO of TargED said:”The first results of Microlyse based on the work of Coen Maas and Stevende Maat underscore the potential of Microlyse to break down blood clots in various diseases. We appreciate the support from our investors to realizenext development steps, and I am honored to be part of a company that has the potential to positively impact millions of lives by enabling patient access to these lifesaving treatments.”

Bart van der Worp, MD PhD,Neurologist specialized in cerebrovascular diseases at the University Medical Centerin Utrecht, stated: “Given its mode of action, Microlyse has the potential not only to improve the chance of recanalization of an occluded artery as compared to intravenous thrombolysisusing currently available drugs, but also to reduce the risk of intracranial bleeding, the most feared complication of thrombolysis. Greater efficacy and an improved safety profile could lead to better outcomes for thousands of patients worldwide. To assess the potential benefit of Microlyse, further preclinical and clinical research is needed.”

Rianne Ellenbroek,PhD,Investment Manager of FIRST commented: “We areveryexcited to support the TargED team ontheir journey to further develop the strong scientific work Coen Maas and Steven de Maat have established with support of the Dutch CardioVascular Alliance(DCVA). Microlyse is a therapeutic innovation that has a strongpotential tolowerthe burden of cardiovascular diseases related to thrombotic events.”

UNIIQ invests €200,000 in novel cancer therapeutics startup BIMINI Biotech

BIMINI Biotech, an innovative therapeutics startup that develops new cancer drugs, has secured an investment of €200,000 from proof-of-concept fund UNIIQ to further develop its technology. The company aims to use the growth hormone receptor to uniquely inhibit cancer cell growth. The investment was announced digitally by Ida Haisma, director of Leiden Bio Science Park.

BIMINI’s technology is based on pioneering research by Professor Ger Strous at UMC Utrecht and has shown promising results in the preclinical phase. The startup recently moved to Leiden to continue preclinical research and make the transition to the clinical phase.

Human growth hormone (GH) modulates cellular metabolism, leading to cell growth, cell reproduction and cell regeneration. GH also stimulates the production and secretion of insulin-like growth factor 1 (IGF-1) in many tissues. GH and IGF-1 both promote cell proliferation and suppress apoptosis.

The scientific literature and numerous animal studies and retrospective human studies indicate that this GH/IGF-1 axis plays an important role in cancer development. Overexpression of the axis in various types of cancer leads to explosive cellular growth, with cells triggering growth in themselves or surrounding cells. The GH/IGF-1 axis is therefore the ideal target for anti-cancer drug development.

Using this axis as a target, BIMINI’s strategy is based on a new mechanism that renders the growth hormone receptor dysfunctional. This approach elegantly mimics nature by making cells less sensitive to growth hormone as seen in Laron syndrome (a specific type of dwarfism). People with this condition, characterised by growth hormone receptor deficiency, have been found to be resistant to diseases like cancer.

Three small molecules (BM001-003) have been identified and patented by BIMINI, of which BM001 is the lead compound currently being used in the preclinical phase. This compound has shown, both in vitro and in vivo, promising anti-cancer effects for different types of cancer.

The GH/IGF-1 axis is not only involved in cancer development but in other areas too, including metabolic disorders and the ageing process. This innovative approach may therefore prove applicable to other indications in the future. Indeed, BIMINI aims to develop a drug discovery platform around the GH/IGF-1 axis and aspires to become a leading innovator in growth hormone receptor-based therapeutics.

The startup will use UNIIQ’s capital injection for additional in vivo efficacy studies as well as for toxicological and ADME experiments, while further strengthening its IP position and organisational development.

Digvijay Gahtory, CEO of BIMINI Biotech: “We can use UNIIQ’s investment to conduct experiments that will be essential for bridging the gap to Series A funding and thus bring our innovative technology one step closer to patients.”

Hans Dreijklufft, fund manager at UNIIQ: “To improve prognosis and life expectancy, it is fundamental that new therapeutics reach cancer patients. BIMINI’s innovative technology has the potential to make a big difference. We are therefore very happy to invest in the company. We have a lot of faith in its team and are convinced that BIMINI Biotech is a strong addition to the Leiden ecosystem.”

European Commission approves MenQuadfi®, the latest innovation in meningococcal (MenACWY) vaccination for individuals 12 months of age and older

The European Commission (EC) has approved MenQuadfi® for active immunization of individuals from the age of 12 months and older against invasive meningococcal disease caused by Neisseria meningitidis serogroups A, C, W and Y.9

“Meningococcal meningitis can take one’s life in as little as one day and leave survivors with severe permanent disabilities.10,11 In Europe, there were more than 3,000 cases of Invasive Meningococcal Disease in 2018, half of them caused by serogroups C, W and Y,”8 says Thomas Triomphe, Head of Sanofi Pasteur. “One case is one too many. It is our ambition to make this vaccine available worldwide to further expand protection to as many people as possible. The European Commission’s approval of MenQuadfi takes us one step closer to achieving this goal.”

The European Commission’s decision is based upon results from a robust and comprehensive international clinical program, including seven pivotal Phase 2 and 3 randomized, active-controlled, multi-center studies. The immunogenicity and safety of MenQuadfi were evaluated in over 6,300 healthy individuals aged 12 months and older, who received a single dose of MenQuadfi.1,2,3,4,5,6,7

MenQuadfi was compared with other licensed combination vaccines across all age groups. It demonstrated a good safety profile and induced a high immune response against all four serogroups (A, C, W and Y) consistently across all studies.1,2,3,4,5,6,7

“The introduction of a new vaccine against four of the major serogroups of meningococcal disease is very welcome news. The disease is unpredictable and remains the biggest cause of sepsis and septic shock in children across Europe today,12” says Professor Federico Martinón-Torres, Pediatrician and Clinical Researcher, Head of Pediatrics and Vaccine Research Unit at Hospital Clínico Universitario de Santiago in Spain. “Meningococcal disease is vaccine-preventable but, in spite of its threat, there is currently no common immunization schedule for it in Europe. The approval of MenQuadfi in Europe will contribute to our efforts to protect against, and help defeat, this truly devastating disease.”

In order to better address the global need for meningococcal disease prevention over the life course, Phase 3 studies are ongoing to investigate the vaccine in infants from 6 weeks of age.13,14,15,16,17,18

Invasive meningococcal disease (IMD) epidemiology is highly unpredictable and varies widely across geographies and over time. In Europe, with the increase in incidence of IMD caused by hypervirulent serogroup W, several countries have introduced MenACWY conjugate vaccination into their routine vaccination schedules. However, considerable variation remains between European countries, leaving room for outbreaks in unprotected and vulnerable populations.19

In 2018, 3,233 individuals contracted invasive meningococcal disease in Europe, and approximately 1 in 10 did not survive. Of the total number of cases, 2,911 were reported to be serogroups B, C, W or Y, of which almost half (47%) were serogroup C, W or Y.8 Rates were highest in infants, followed by children under 5 years, with a second peak in those aged 15–24 years.8

Selvita to acquire Fidelta from Galapagos

Galapagos NV [Euronext & NASDAQ: GLPG] and Selvita S.A. [WSE: SLV] – announced today that they have signed an agreement under which Selvita will acquire 100% of the outstanding shares in Fidelta d.o.o. for an enterprise value of € 31.2M plus the customary adjustments for net cash and working capital.

Fidelta is a contract research organization within the Galapagos Group of companies, with core scientific competences in inflammation, fibrosis, and anti-infectives. It currently employs 181 employees, including over 150 highly experienced scientists, providing integrated drug discovery services in the biotech and pharmaceutical industry, with a proven track record of accomplished laboratory projects over many years. Fidelta is located in state-of-the art R&D facilities in Zagreb, Croatia which offer almost 6,000 m2 of research space. The transaction is subject to customary closing conditions and is expected to close on the 4th of January 2021.

Galapagos is a fully integrated biopharmaceutical company with a deep pipeline of novel mechanism of action candidates and commercial operations in Europe; as such, the CRO activities offered by Fidelta no longer fit with its strategy.

Upon completion of the acquisition, Fidelta will be fully consolidated under the Selvita Group, and will continue to operate under the Fidelta name.

The scope of services provided by Fidelta is complementary to Selvita’s offerings and will enable Selvita to build a competitive advantage in business areas such as DMPK, in vivo pharmacology, and toxicology. Selvita will also increase its scale of operations within medicinal chemistry and in vitro pharmacology, resulting in significant strengthening of its market position.

Fidelta’s therapeutic areas of expertise align with current market trends, as infectious diseases, inflammation, and fibrosis are all areas of core expertise for Fidelta and are becoming increasingly major areas of interest for pharma companies due to the significant and unmet medical needs.

“We are thrilled to announce our acquisition of Fidelta. This is a perfect fit for us: an extensively experienced organization, with a full range of in-house drug discovery capabilities and an established track record of clinical candidate delivery,” says Boguslaw Sieczkowski, Chief Executive Officer at Selvita. “With this acquisition, we are not only executing our strategy announced this year, but we’re actually taking a leap in both the scope of our services portfolio and the scale of business. It is a transformative moment in the history of Selvita.”

“The scientific knowledge, the business acumen, and the focus and commitment of the team at Fidelta are second to none.  I would like to thank the Fidelta team for the excellent collaboration over the years.  I’m sure they will thrive under the Selvita umbrella. We will continue part of our discovery work with Fidelta, so we will benefit from that,” says Onno van de Stolpe, CEO of Galapagos.

“We are very happy to take this big and logical step in the further development of Fidelta, and together with Selvita, to strengthen our presence on the market. Bringing together these two companies will allow us to keep a high level of quality and offer an even more integrated approach  in services that we provide to our clients,” says Adrijana Vinter, Managing Director of Fidelta.
