InteRNA Technologies announced today that the first patient has been dosed in the first cohort of its first-in-human Phase I study with the Company’s lead microRNA candidate, INT-1B3. The trial will evaluate safety and initial signs of efficacy of INT-1B3, a microRNA-mimic of the endogenous tumor suppressor miR-193a-3p formulated in next-generation lipid nanoparticles, in patients with advanced solid tumors. INT-1B3 represents a promising novel therapeutic approach that could address multiple hallmarks of cancer simultaneously by directly targeting tumor cells and the tumor microenvironment by specific modulation of multiple relevant signaling pathway components triggering a long-term T cell-mediated immune response against the tumor.
“The enrollment of the first patient in this trial, especially during the ongoing COVID-19 pandemic, is a significant milestone for InteRNA,” said Dr. Roel Schaapveld, CEO of InteRNA Technologies. “microRNAs represent a new class of therapeutic agents that have the potential to change the treatment paradigm in immune-oncology, delivering a combination approach as a single regimen. We are very pleased that the first patient completed the first cycle without dose-limiting toxicity, and enrolment of the second cohort could already be initiated. The trial will allow us to generate further important insights on the potential and underlying mechanisms of INT-1B3 and reinforces our commitment to bringing this novel modality to patients, especially for the treatment of hard-to-treat solid tumors.”
This two-part, open-label, multiple ascending dose Phase I/Ib trial (NCT04675996) will evaluate the safety, pharmacokinetics, pharmacodynamics, and preliminary efficacy of INT-1B3. The dose escalation part of the study (part 1) will be conducted in trial sites located in the Netherlands and Belgium and will enroll approximately 30 patients with advanced solid tumors. The dose expansion part of the trial (part 2) will be conducted in multiple clinical study centers located in several European countries as well as in the United States and will enroll up to 50 patients with hepatocellular carcinoma or triple negative breast cancer. All patients will receive INT-1B3 via infusions twice per week in 21-day cycles. Topline results from the dose escalation part of the study are expected by the end of 2021.

Vico Therapeutics,a Leiden, the Netherlands, based biotech company focusing on the development of RNA modulating therapies for rare neurological disorders, announced that the European Commission (EC) has granted orphan drug designation for VO659, Vico’s investigational antisense oligonucleotide therapy for the treatment of Spinocerebellar Ataxia (SCA). The orphan designation was based on a positive opinion from the European Medicines Agency (EMA) Committee for Orphan Medicinal Products (COMP). Vico previously received orphan drug designation for VO659 in Huntington Disease.
“Spinocerebellar Ataxias make up a great part of the so called polyglutamine disorders, debilitating and progressive diseases, leading to significant impairment of mobility, speech, and multiple other daily activities of patients suffering from this condition. So far, there is no treatment which can halt the progression of these diseases,” said Rupert Sandbrink M.D. Ph.D., Chief Medical Officer at Vico.
“Our investigational RNA modulating therapy is designed to lower the mutant protein levels causing these neurodegenerative diseases. We’re pleased to receive orphan drug designation, which recognizes both the unmet medical need for patients living with spinocerebellar ataxias, and the potential of our approach.”
Qualified for orphan drug designation are drugs and biologics intended for the safe and effective treatment, diagnosis or prevention of rare diseases or conditions that impact fewer than 5 in 10,000 patients in the European Union. Orphan drug designation gives developing companies certain benefits, including clinical protocol assistance, reduced regulatory fees, research grants and 10 years of market exclusivity following regulatory approval.

Forbion, a leading European life sciences venture capital firm, today announces the launch of VectorY, a fully integrated gene therapy company focused on the development of innovative vectorized antibodies for muscular and neurodegenerative disorders. The Company has been operational since October 2020 and has established laboratories and offices at the Amsterdam Science Park.
VectorY develops proprietary and partnered programs based on a novel AAV platform and innovative antibody-based targeted degradation technologies. The Company is creating a pipeline of innovative vectorized antibodies targeting muscular and CNS diseases, aimed at overcoming limitations of current therapies, improving delivery, durability and accessibility of targeted tissues and cells. These novel viral vectors will have enhanced properties of non-immunogenicity, cell type specificity and expression in selected sub-cellular compartments, thus unlocking new treatment opportunities in these disease areas.
This year the Company is setting up its own state-of-the-art GMP manufacturing facilities in Amsterdam. Its technology platform will be developed for scalability and yield, with the aim of significantly lowering the costs of gene therapies.
VectorY has already negotiated its first corporate collaboration agreement, gaining access to a highly relevant neuro-inflammation target to develop a vectorized antibody therapy for treatment of neurological disorders. VectorY will continue to consider further collaborations in parallel to progressing its proprietary programs.
The highly experienced management team will be led by Marco Boorsma, General Partner at Forbion, who is joining as Interim CEO, bringing a proven track record of building and leading biotech companies. Co-founders Carlo Incerti, Pavlina Konstantinova, Anthony Newcombe and Barbara Sanders all bring significant, longstanding experience in gene therapy, vector development, and biopharmaceutical manufacturing expertise. Carlo Incerti, Operating Partner at Forbion and previously Chief Medical Officer of Sanofi Genzyme, will join VectorY’s board as Chairman, together with Sander Slootweg, Managing Partner at Forbion, as board member. Forbion’s principal Dmitrij Hristodorov will join as Observer to the board.
VectorY was seeded by Forbion, following previous investments in successful gene vector companies including bluebird bio (then named Genetix), Biovex, Hookipa, EnGene, Amsterdam Molecular Therapeutics, uniQure and Replimune.
Carlo Incerti, Co-founder and Chairman of the Board of VectorY, and Operating Partner at Forbion, said: “I have been working in the gene therapy field for over 30 years, and, from experience, know that accurately targeting the right cells with the transgene has always been of paramount importance. Using viral vectors developed at VectorY to deliver the genetic sequence for therapeutic antibody expression in target cells has the potential to address some of the key shortcomings of intravenously injected molecules. Our aim is to create novel treatments for patients.“
Marco Boorsma, Interim CEO of VectorY, said: “VectorY is developing innovative technologies to create novel, potentiated vectorized antibodies that will target muscle and CNS disorders with substantial unmet medical needs. Our aim is to bring new, powerful treatments to patients suffering from serious diseases. The Company is hitting the ground running, capitalizing on the extensive experience of its co-founders, and the support and expertise from Forbion as longstanding gene therapy investor.”

The biotech sector in the Netherlands [has] grown up the last few years,” zegt onze eigen Annemiek Verkamman. “A couple of years ago, a lot of people were in early phases, maybe clinical Phase 1. And now we see the first Dutch biotechs really deliver a product to the market, which is of course a real milestone.”
“Now, the Netherlands is on the map”, zo concludeert HollandBIO’s Annemiek in het artikel. Of, zoals de journalist het samenvat: “Biopharma Looks to the Netherlands as European Hub”.

InteRNA Technologies announced today the closing of an extended Series B financing round amounting to EUR 18.5M in total. This Series B round was led by AurorA Science, an Italian biotech investment company, along with existing investor Waterman Ventures. Current shareholders Aglaia Oncology Funds and OostNL also contributed to the financing round. The funding will enable the clinical evaluation of the Company’s microRNA lead candidate, INT-1B3, in patients with advanced solid tumors. Furthermore, the proceeds will be used to develop and advance additional proprietary preclinical drug candidates adressing a variety of cancer indications thereby expanding the Company’s pipeline. As part of the Series B financing, Gabriele Campi, PhD, of AurorA Science will join the Board of Directors.
“This successful extension of the Series B financing round validates the unique potential that microRNAs offer as a therapeutic modality for hard-to-treat cancers,” said Dr. Roel Schaapveld, CEO of InteRNA Technologies. “We thank AurorA Science, Waterman Ventures and our other long-term investors for their support in bringing our microRNA technology into the clinic and look forward to evaluate INT-1B3’s unique mode of action, targeting not only the tumor cells themselves but also the disease-promoting tumor microenvironment.”
“With this participation in InteRNA Technologies, AurorA Science continues its strong commitment into front-edge science,” commented Guido Guidi, Chairman at AurorA Science. “We believe InteRNA has developed a promising innovative technology in this area and look forward to supporting the team on their path forward.” microRNAs are naturally occurring, non-coding strands of RNA that trigger the RNA interference pathway and regulate gene expression by controlling the efficiency of messenger RNA (mRNA) translation into functional proteins. InteRNA designs breakthrough therapies tackling cancer by using the inherent characteristic of microRNAs to simultaneously inhibit multiple mRNA targets in a coordinated fashion, e.g. by blocking activated cancer signaling pathways and preventing activation of alternative disease-promoting pathways.

InteRNA Technologies Extends Series B Financing Round Totaling EUR 18.5M
InnovatieklimaatInteRNA Technologies announced today the closing of an extended Series B financing round amounting to EUR 18.5M in total.
