Genmab Enters Strategic Partnership with Immatics to Discover and Develop Next Generation Bispecific Cancer Immunotherapies

Genmab has entered into a research collaboration and exclusive license agreement with privately owned Immatics Biotechnologies GmbH (Immatics), to discover and develop next-generation bispecific immunotherapies to target multiple cancer indications. The deal strengthens Genmab’s position in immuno-oncology by combining Genmab’s proprietary technologies and antibody know-how with Immatics’ XPRESIDENT® targets and T-cell receptor (TCR) capabilities.  Genmab will receive an exclusive license to three proprietary targets from Immatics, with an option to license up to two additional targets at predetermined economics. The companies will conduct joint research, funded by Genmab, on multiple antibody and/or TCR-based bispecific therapeutic product concepts.

Genmab may elect to progress any resulting product candidates, and will be responsible for development, manufacturing and worldwide commercialization. For any products that are commercialized by Genmab, Immatics will have an option to limited co-promotion efforts in selected countries in the EU.

Cleara Biotech Announces Three Collaborations to Develop New Therapeutics Targeting Senescent Cells and Cancer

Cleara Biotech, a Netherlands-based biopharmaceutical company, announced today the creation of three public-private partnerships to discover and develop new therapeutics. Cleara has partnered with Dr. Peter de Keizer from the University Medical Center in Utrecht, Dr. Marco Demaria from the University Medical Center in Groningen, and Dr. Tobias Madl from the Medical University of Graz to engage in its discovery operations. Cleara’s initial funding was led by founding investor Apollo Ventures, a life sciences venture capital firm and company builder working across Europe and North America.

Cleara is a preclinical R&D company engaged in the creation of therapeutics targeting the unique biology of senescent cells to treat age-related pathologies and therapy-resistant cancer. Senescent cells are rare cells that accumulate in our bodies as we age and contribute to the aging process. Removing these cells has been shown to reverse signs of aging including muscle and hair loss, cancer development, energy levels, and even increase healthy lifespan of mice.

Cellular senescence is a remarkably new field of study, and some of the world’s top senescent cell researchers have come together in Cleara to translate their scientific work on mice into human therapeutics. “These partnerships of top-notch academic institutions with drug development efforts will allow us to accelerate clinical development of our lead programs and provides an opportunity to treat multiple diseases by eliminating a patient’s senescent cells,” said Dr. de Keizer.

EUROSTARS grants project funding for IMMUNOMONITOR consortium led by ENPICOM

A total grant sum of EUR 2 million has been awarded to a multidisciplinary EUROSTARS consortium. The EUROSTARS project is titled “Treatment Response Monitoring for Cancer Immunotherapies Using Immune Repertoire Analysis” and named IMMUNOMONITOR.

This European consortium consists of five collaborating partners: ENPICOM (Netherlands), a bioinformatics and software engineering company; Biomax Informatics (Germany), a knowledge management solution company; Ultimovacs (Norway), a pharmaceutical company developing innovative vaccines for treatment of cancer; the lab of Prof. Binder of the University Medical Center Hamburg-Eppendorf or UKE (Germany); and OSE Immunotherapeutics (France), a biotechnology company developing innovative immunotherapies for immune activation and regulation in the fields of immuno-oncology and autoimmune diseases.

ENPICOM develops a T-cell/B-cell receptor (TCR/BCR) repertoire immunosequencing data analysis platform to support the development, patient stratification and treatment monitoring of immunotherapies. The goal of the IMMUNOMONITOR project is to validate the data analysis and visualization software solution for treatment response monitoring of cancer vaccines based on a unique approach to over-time immune repertoire sequencing data analysis.

While a growing number of immunotherapies have been approved, no adequate diagnostic tests are available to capture the desired immune response and to monitor if the therapy is effective in an individual patient. The consortium is addressing this unmet medical need with a software solution to capture and monitor a patient’s dynamic immune response over time using blood samples. Advances in Next-Generation Sequencing (NGS) allow taking ‘snapshots’ of a patient’s immune response by sequencing DNA or RNA from the patient’s immune cells (i.e. immune repertoire). The enormous diversity of immune repertoires results in a staggering amount of data, which cannot be handled by existing tools for NGS data analysis.

In the IMMUNOMONITOR project, advanced bioinformatics algorithms will be developed using NGS big data analysis to generate an ‘immune status fingerprint’ for monitoring cancer vaccines. The clinical validation studies will include two cancer vaccines tested in pancreatic cancer, lung cancer and malignant melanoma developed by OSE Immunotherapeutics and Ultimovacs, respectively.

De opmars van fotonica: ‘1000 nieuwe banen voor Twente’

PhiX, een dochterbedrijf van LioniX, heeft een machine voor de assemblage van zogeheten ‘fotonica-chips’ in gebruik genomen. Het Enschedese bedrijf moet de komende zeven jaar uitgroeien tot een onderneming met 1000 werknemers.

Fotonica-chips zijn bezig aan een snelle opmars. De nieuwe generatie chips wordt onder meer gebruikt in de nieuwe standaard voor mobiele technologie 5G, in medische apparatuur en in systemen voor de opslag van data. De nieuwe chips zijn energiezuinig en snel. PhiX is het eerste bedrijf ter wereld, dat deze nieuwe chips geautomatiseerd kan assembleren. Daarbij worden glasvezels die tien keer dunner zijn dan een haar verlijmd. Dat proces duurt enkele seconden, terwijl het een uur tot anderhalf uur per chip duurt, wanneer het met de hand wordt gedaan. De nieuwe machine is in samenwerking met het Fraunhofer Projectcenter ontwikkeld. Fraunhofer is de Duitse tegenhanger van TNO en vestigde zich anderhalf jaar geleden op de UT. Het patent voor de nieuwe machine wordt gedeeld door Fraunhofer en Phix.

Twente loopt voorop

Phix is een dochterbedrijf van LioniX in Enschede (80 werknemers), dat de fotonica-chips produceert. De assemblage gebeurt bij LioniX nu dus nog handmatig. Fotonicachips zorgen volgens operationeel directeur Joost van Kerkhof van Phix voor een technologische revolutie. „We lopen met deze technologie in Twente voorop.” Naast moederbedrijf LioniX investeerden ook Oost NL, de UT, Saxion en enkele particuliere investeerders in PhiX. Ze zorgden voor de eerste 1,2 miljoen euro en volgende maand volgt een tweede financieringsronde in dezelfde orde van grootte.

120 miljoen nodig

Directeur Hans van den Vlekkert  van LioniX is ervan overtuigd dat een omzet van 1 miljard euro voor LioniX en Phix samen in 2025 haalbaar is. Daarvoor moet er dan wel een nieuwe fabriek worden gebouwd. Realisatie van een nieuwe chipfabriek (LioniX) kost 80 miljoen en een assemblagefabriek (PhiX) 40 miljoen. Er liggen volgens Van den Vlekkert ook aanbiedingen uit China en Zuid-Korea om daar de fotonica-chips te laten maken. „Voor de hand ligt dat we de chips eerst in het Verre Oosten laten produceren en de assemblage hier in Twente gaan doen. Als er investeerders voor te vinden zijn, tenminste. De assemblage kan 900 tot 1000 nieuwe banen opleveren bij PhiX.”

MKB-Actieplan mooie opsteker voor biotech sector

Vorige week presenteerde staatssecretaris Mona Keijzer de plannen van de overheid om het MKB te stimuleren. Het kabinet maakt daar tot en met 2021 in totaal 200 miljoen euro extra voor beschikbaar. Eén van de belangrijkste onderdelen van dit MKB-Actieplan is de verruiming van het budget van het Innovatiekrediet van 2019 tot en met 2021. HollandBIO is blij dat er na onze jarenlange en intensieve inspanningen eindelijk sprake is van een structurele toename. Het is na de verhoging van 2018 opnieuw een mooi resultaat voor de biotech sector, en een goede stap in de richting van een ijzersterk innovatieklimaat.

 

In het eerste jaar als staatssecretaris bezocht Mona Keijzer diverse MKB-ondernemingen tijdens de MKB-tour, waaronder biotech bedrijven op het Pivot Park en Leiden Bioscience Park. Hierbij besprak zij de drie grootste uitdagingen voor het MKB: personeel, digitalisering en financiering. Met name voor financiering pakken de aankondigingen erg positief uit. Het actieplan maakt ruim 120 miljoen euro vrij voor het behouden én intensiveren van de huidige regelingen. Naast uitbreiding van innovatie-instrumenten als het Innovatiekrediet, de MIT-regeling en het SBIR-programma, gaat er ook meer geld naar de SEED-regeling en komt er meer aandacht voor human capital. Verder komt er een regeling voor Thematische Technology Transfer om kennisinstellingen die excellent onderzoek verrichten op bepaalde thema’s met elkaar te verbinden voor een betere kennisbenutting.

 

Een ijzersterk innovatieklimaat

De plannen zijn aanvullend aan de nieuw op te zetten financieringsinstelling Invest-NL. Met Invest-NL komt er een platform dat kan dienen als vliegwiel voor de doorgroei van biotech bedrijven in verschillende fases van ontwikkeling. Het MKB-Actieplan waarborgt én versterkt de bestaande stimuleringsmaatregelen voor het MKB. Samen dragen zij bij aan een uitgekiende financieringsmix voor bedrijven in de life sciences. HollandBIO blijft zich inzetten voor een ijzersterk innovatieklimaat in Nederland. Zo blijven wij ook in de toekomst genieten van oplossingen uit de biotech die bijdragen aan een duurzame en gezonde samenleving.

VarmX haalt € 12,5 miljoen aan financiering op

VarmX heeft in totaal € 12,5 miljoen aan nieuwe financiering opgehaald. Deze financiering is bestemd voor de ontwikkeling van gemodificeerd bloedstollingsfactor X voor de behandeling van ernstige bloedingen bij patiënten die antistollingsmiddelen gebruiken.

 

VarmX had eerst een Serie A-financieringsronde van € 7,5 miljoen afgesloten. De ronde werd geleid door aandeelhouder BioGeneration Ventures (BGV), met daarnaast medeaandeelhouder InnovationQuarter Capital, de economische ontwikkelingsonderneming voor Zuid-Holland. Ook investeringfonds Uniiq en de Universiteit Leiden zijn aandeelhouders door de conversie van hun starterslening in VarmX. De financiering wordt aangevuld door de Rijksdienst voor Ondernemend Nederland (RVO.nl) met een innovatiekrediet van € 5,0 miljoen tot een totaal van € 12,5 miljoen. Dit werd dit weekend bekendgemaakt op het Dutch Biotech Event van HollandBIO in Maarssen. De Serie A-ronde volgt op de initiële startfinanciering in juni 2017 door BGV en InnovationQuarter.

 

VarmX ontwikkelt PseudoXa – dat is gebaseerd op onderzoek uitgevoerd door het LUMC naar de eigenschappen van slangengif en van menselijke bloedstollingsfactor X – voor het stoppen of voorkomen van bloedingen bij patiënten die antistollingsmiddelen gebruiken in de vorm van synthetische factor Xa-remmers. De nieuwe financiering zal VarmX in staat stellen om de ontwikkeling van PseudoXa als veilig en effectief geneesmiddel te versnellen en vroege klinische studies mogelijk te maken.

 

Om beroerte of diepe veneuze trombose (trombosebeen) te voorkomen, nemen wereldwijd miljoenen patiënten dagelijks synthetische factor Xa-remmers, zoals apixaban, edoxaban of rivaroxaban. Het gebruik van deze geneesmiddelen neemt snel toe. Als bijwerking treedt ieder jaar bij 2-3% van de patiënten die deze antistollingsmiddelen gebruiken een ernstige (interne) bloeding op die moeilijk te behandelen of te voorkomen is bij een spoedoperatie. PseudoXa biedt een veilige en effectieve oplossing voor dit bloedingsprobleem door het vermogen om bloedstolling onmiddellijk te herstellen ondanks de aanwezigheid van factor Xa-antistollingsmiddelen.

 

Impuls richting kliniek

Met hun Serie-A investering, na startfinanciering van vorig jaar, stellen BGV en InnovationQuarter, samen met RVO.nl, VarmX in staat om het productieproces te ontwikkelen en verder op te schalen, met de nadruk op zuivering en formulering en op de farmacodynamiek en -kinetiek van de werkzame stof. Daarnaast is alles gericht op het aantonen van veiligheid in preklinische modellen en in de mens. Er zal ook worden gewerkt aan de ontwikkeling van nieuwe therapieën op het gebied van bloedstolling , die VarmX in de nabije toekomst zullen helpen bij de doorgroei tot een volwassen biotechnologische onderneming met een pijplijn aan producten.

AbbVie dumps Galapagos’ cystic fibrosis combo

AbbVie has dumped one of Galapagos’ cystic fibrosis regimens after getting a look at underwhelming clinical data on a component of the triple combination. The action prompted Galapagos to review the future of its relationship with AbbVie—and provided a boost to cystic fibrosis rival Vertex.

Galapagos has moved one AbbVie-partnered triple combination into clinical development but hopes are higher for the follow-up regimen, which features a potentiator with a shorter metabolite half-life. Despite that, AbbVie has decided against proceeding with the second, more-promising combination after reviewing data on the C2 corrector, GLPG2737, that features in both regimens.

Investigators enrolled 22 cystic fibrosis patients who were homozygous for Class II F508del mutation and on stable treatment with Vertex’s Orkambi. Two-thirds of participants received GLPG2737 on top of Orkambi, while the rest received placebo plus Vertex’s drug.

The trial met its primary, sweat chloride concentration endpoint but failed against a measure of lung function. Notably, the percentage change in the ppFEV1 measure of lung function—3.4%—came in below the 5% targeted by analysts and well under the 7% to 13% improvements achieved by Vertex’s triple combinations. Using a higher dose of GLPG2737 and pairing it with Galapagos’ drugs, not Orkambi, could give the triple combinations better efficacy but for now Vertex looks to have an edge.

Vertex’s combinations are further down the pipeline, too, meaning Galapagos faces the prospect of trying to come from behind with what may prove to be a less efficacious therapy.

“The key is Vertex sees robust and rapid effects with all of their combos and is already well into phase 3 with at least two—and Galapagos has not put out much triple data thus far,” Jefferies analyst Michael Yee wrote in a note to investors.

Galapagos may have to tackle the challenge without AbbVie. For now, AbbVie remains partnered on the first triple combination. But Galapagos is reviewing the broader cystic fibrosis partnership in light of AbbVie’s decision to drop the second triple combination.

AbbVie’s decision further complicates one of the more fractious partnerships in biotech. Three years ago, AbbVie dumped Galapagos’ JAK inhibitor filgotinib and switched focus to its rival drug, leaving its one-time partner trailing in the race to market. The existing cystic fibrosis partnership continued but there have been signs of tension. According to Dutch newspaper Het Financieele Dagblad, Galapagos CEO Onno van de Stolpe thinks AbbVie is to blame for delays to the cystic fibrosis program and that it would advance just as well without the Big Pharma.

Galapagos may now get a chance to find out whether van de Stolpe is right.

Wouter Bos wordt topman Invest-NL

Wouter Bos, bestuursvoorzitter van het VUmc en oud-minister van Financiën, wordt de eerste bestuursvoorzitter van Invest-NL, de staatsdeelneming die investeringen gaat stimuleren op maatschappelijke terreinen waar Nederland nu nog kansen laat liggen. Invest-NL zal naar verwachting in 2019 definitief worden opgericht en ondernemers dan één loket bieden voor risicokapitaal, garanties en financieringsprogramma’s.

In aanloop naar de oprichting zal Bos eind oktober van dit jaar beginnen met de opbouw van Invest-NL om zodoende een voortvarende start te kunnen maken zodra de oprichting van de naamloze vennootschap een feit is. Bos krijgt in deze opbouwfase de gelegenheid om de strategie en visie van Invest-NL mede vorm te geven.

Minister van Financiën Wopke Hoekstra, minister van Economische Zaken en Klimaat Eric Wiebes en minister voor Buitenlandse Handel en Ontwikkelingssamenwerking Sigrid Kaag zijn verheugd over de aanstelling van de heer Bos. Hij combineert bestuurservaring in zowel de private als de publieke sector met een sterke betrokkenheid bij de maatschappelijke thema’s waar Invest-NL zich op zal richten.

Het initiatief voor Invest-NL werd in 2017 genomen door het vorige kabinet. Invest-NL zal ondernemers ondersteunen bij grote maatschappelijke projecten en waar nodig meefinancieren. Dankzij Invest-NL is er straks méér geld om méér te doen voor méér ondernemers. Deze investeringen zijn nodig om onze economie verder te versterken en de omslag naar een duurzame economie te maken. Invest-NL gaat daarbij helpen.

Nu is het nog zo dat veel bedrijven en projecten vanwege onzekerheid over de verhouding tussen risico en rendement of door lange, onzekere terugverdientijden op investeringen, moeilijk aan financiering komen. Invest-NL gaat dat probleem verhelpen met het bestaande financieringsinstrumentarium en met de kapitaalstorting van 2,5 miljard euro. Daarmee zal ook meer financiering vanuit institutionele beleggers en Europese fondsen en programma’s worden aangetrokken.

De instelling gaat daarnaast ook start-ups en scale-ups helpen. Deze innovatieve bedrijven bevinden zich nog in het beginstadium maar hebben kapitaal nodig voor verdere groei. Invest-NL kan durfkapitaal leveren door bijvoorbeeld een belang te nemen in een fonds dat investeert in dergelijke bedrijven. Ook kan Invest-NL straks zelf participeren in doorgroeiende bedrijven.

De ministerraad heeft vrijdag ingestemd met het wetsvoorstel voor de oprichting van Invest-NL. Minister Wiebes van Economische Zaken en Klimaat zal het wetsvoorstel voor advies voorleggen aan de Raad van State.

Trend analysis: Great year for Dutch biotech R&D

Today, the leading global business development database Biotechgate presents its 2018 trend report for the Dutch life sciences market. The annual trend report confirms the top position of the Netherlands in the life sciences industry.

For the Dutch biotech research and development efforts, 2017 proved to be remarkably fruitful. The development pipeline contains more products than previous years. Currently, 42 phase I, 47 phase II and 11 phase III studies are registered, which is almost twice as much as the total number of studies reported two years ago. And the preclinical studies are impressive, too. In total, 97 products are in preclinical development, holding promise for a bright future.

The total number of biotech companies is relatively stable: 455. The analysis reflects a well-established Dutch biotech sector. The data confirms the status of The Netherlands as Europe’s most attractive and innovative biopharmaceutical industry environment.
